Foot and Mouth Disease : Etiology, Epidemiology and Control Measures

Foot and mouth disease (FMD) is highly infectious disease of cloven-hoofed animals, particularly cattle, sheep, pigs and goats. Also, it is the most important animal pathogen on the global scale because of the potential for rapid and extensive spread through susceptible animal populations. Outbreak can lead to formidable economic consequence for domestic livestock production and international trade. more » ... the genus Aphthovirus within the family Picornaviridae. There are seven immunologically distinct serotypes; O, A, C, SAT (Southern African Territories) 1, SAT 2, SAT 3 and Asia 1 and a diverse antigenic spectrum of virus strains within each serotype. Characteristic lesion of FMD is the formation of vesicles in the mucosal membranes of mouth, muzzle, foot, and teats. Nowadays, many developed countries have maintained FMD-free as a result of eradication efforts. However, outbreaks of FMD have occurred in several countries, even in Europe, and it is still endemic in Africa, the Middle East, Asia, and South America. Last year, three outbreaks of FMD occurred in our country. Last outbreak reported in November, 2010 induced the enormous social and economical impacts. Culling of infected animals, movement control, and vaccination are the major control measures of FMD. To control the disease, each country has their own strategies based on the current situation of FMD in their country. Therefore, I would like to discuss the causative agent, epidemiological properties and control measures of FMD in this paper. 서론 구제역(Foot and mouth disease)은 소, 돼지, 양, 흑염소, 사슴 등의 우제류 동물에 감염될수 있는 전염성이 매우 높은 질병이며, 구제역이 발생하면 살처분 등 질병방제 비용, 생산성감소, 동물 및 축산물의 국제적인 교역 제한 등으로 막대한 경제적인 피해 가 발생한다[1-5]. 원인체인 구제역 바이러스는 Picornaviridae 속의 Aphthovirus 과 에 속하는 envelope 가 없는 이십면체(icosahedral)의 작은 RNA virus 이다[6, 7]. 구 제역은 임상증상으로 구강상피세포, 유방, 콧등, 발굽의 coronary band에 수포를 형 구제역 바이러스는 Picornaviridae, Aphthovirus 속에 속하며 크 기가 가장 작은(23+2 nm)바이러스중 하나로서 single-strand RNA (8 kb)의 핵산을 가지며, 외피가 없고, VP1, VP2, VP3, VP4 구조단백질 (structural proteins)과 여러종류의 비구조 단백질(non-structural proteins) 으로 구성된 icosahedral 형태의 바이러스로서, 상피세포 에 높은 친화성을 가지고 있어 상피세포 발아층에서만 증식되는 특성 을 지니고 있다. 이 바이러스는 현재까지 7종의 혈청형(A, O, C, SAT-1, SAT-2, SAT-3, Asia-1)과 80여종의 혈청아형이 밝혀져 있으며, 혈청형 또는 혈청아형내에서 변이가 잘 일어나며[6, 7, 10, 11], 혈청형간의 교차 면역은 이루어지지 않는 것으로 알려져 있다[11]. 구제역 바이러스는 유 기용매, 낮은 온도, 습기 있는 중성 pH의 환경에서 일반적으로 저항성을 나타내나, 열, 산 및 알칼리에 약하며, 특히 pH에 의한 감수성이 매우 높 아 pH가 6.5 이하나 11이상일 때 빠르게 감염력이 소실된다. 그러므로, 중성 pH의 임파절과 골수에서 장기간 생존이 가능하나, 근육에서는 사 후강직 후 pH는 6.0 이하로 변하므로 바이러스가 급격히 파괴된다[12, 13]. 또한 건조에 민감하여 50% 이하로 상대습도가 저하될 경우 급격 히 그 생존력이 상실된다. 일반적으로 냉장 또는 냉동 상태에서는 감염 력이 유지되나, 50℃ 이상의 온도에서는 급격히 불활성화 되며, 온도와 pH 상태에 따라 다르나 일반적으로 오염된 사료에서 1개월 동안 생존할 수 있다[14-16]. 구제역 바이러스는 동물과 육류, 저장온도(냉장, 냉동) 에 따라서 저항성이 다양하여 짧게는 6-8일에서 길게는 210-352일까 지 감염력을 유지할 수 있다[14]. 오염된 환경에서의 바이러스의 생존은 오염된 물질에 따라서 다르게 나타나며, 바이러스의 초기농도가 중요하 나, 바이러스주, 습도, pH, 기온 등에 영향을 받으며, 야외 상황에서는 매 우 다르게 나타날 수 있다. 18-20℃의 소 털에서는 4주, 건조된 분변에 서 14일 까지, 오줌에서 39일까지, 토양에서 여름에는 3일, 가을에는 28 일까지 살아남을 수 있다[11, 17]. 소의 타액에서는 1시간 후에 0.1% (상 대습도 70%)로 생존이 가능하며, 소와 돼지의 위 및 장의 내용물에서 는 생존기간이 3일 이하이다[16, 18]. 구제역 바이러스를 실험적으로 감 염된 동물에서 유래된 내장을 이용한 자연산 케이싱에서 구제역바이러 스가 250일 까지 존재한 보고가 있다[19]. 건초에서는 200일 이상 생존 하며, 사료는 12-20℃에서 52 일간, 2-5℃에서는 70일간 생존한다. 퇴 비에서는 냉장시 66일, 냉동시 168일, 실온에서는 12일, 밀짚(2℃)에서 232일, 초지의 경우 양지에서 수일간, 가을에는 195일, 겨울에는 262일 이상 생존가능하다[16, 18]. 구제역 바이러스는 사람의 코와 후두에서 24-36시간까지 생존할 수 있어[18] 사람으로부터 소 등으로 공기전파 가 가능하기 때문에 감염된 농장이나 실험실 출입자는 출입 후 일주일 이상 감수성동물과 접촉하지 않아야 한다[16, 18]
