Spectrofluorimetric Method for Determination of Sitagliptin Phosphate in Formulation and Spiked Human Urine

A simple, sensitive and reproducible spectrofluorimetric method was developed for the determination of sitagliptin phosphate in bulk, pharmaceutical formulations and human urine. A simple and reliable validated spectrofluorimetric method was developed for the determination of sitagliptin phosphate. The quantification of sitagliptin was carried out a t a fluorescence wavelength of 297 nm. The linearity range of the method is 0.6-10 μg mL −1 .The limit of detection and limit of quantification for
more » ... quantification for the sitagliptin standard solution were found to be 78.782 ng/ml and 238.735 ng/ml respectively. The percentage amount of sitagliptin phosphate in tablet formulation was found in the range of 93.34-102.67%. The results of within day and interday precision in term of %RSD was found to be less than 5% and 8% respectively. The applicability of the method was tested by spiking of urine with known amount of sitagliptin standard, following centrifugation and membrane filtration of urine sample. The developed method can be successfully employed for the determination of sitagliptin phosphate in various pharmaceutical dosage forms and provides basis for further pharmacokinetic study. The developed method can be used for pharmacokinetic application for the determination of sitagliptin phosphate in human urine analysis.
