Determination of the anxiety- related SCC6A4 gene promoter S and L alleles in Turkish Soccer Players

One of the most important members of serotonin metabolism is serotonin transporter (5HTT), which takes the serotonin back from the synaptic cavity into the presynaptic neuron (reuptake). more » ... med to determine the distribution of the short (S) and long (L) alleles of SLC6A4 gene in 44 young, successful and healthy Turkish male soccer players. Genotyping process was completed by conventional polymerase chain reaction (PCR) after DNA isolation. According to the results, 10 (23%) of the players had LL, 25 (57%) had LS and 9 (20%) had SS genotypes. In the allele distributions, L allele was counted as 45 (51%) and S allel as 43 (49%). It has been reported that SLC6A4 alleles show large variation in individuals with neuropsychiatric disorders compared to the healthy individuals. This is the first study to report that LL genotype and L allele are more frequent than SS genotype and S allele in Turkish young professional soccer players. Özet Serotonin metabolizmasının en önemli üyelerinden biri, seratonini sinaptik boşluktan presinaptik nörona geri alan serotonin taşıyıcı proteindir (5HTT). Bu protein SLC6A4 geni tarafından kodlanır ve bu genin promotor bölgesinde ki fonksiyonel delesyon/ insersiyon mutasyonu , genin transkripsiyon hızına etki eder. "S" alleli genin kısa formu olarak adlandırılır ve günümüze kadar yapılan çalışmalar ile kaygı ile ilişkisini gösterilmiştir. Bu çalışmamızda SLC6A4 geninin 44 genç, başarılı ve sağlıklı Türk erkek futbolculardaki kısa (S) ve uzun (L) allel dağılımının belirlenmesi amaçlanmıştır. Genotipleme çalışması DNA izolasyonu sonrasında konvansiyonel polimeraz zincir reaksiyonu (PCR) ile tamamlanmıştır. Analiz sonuçlarına göre futbolcuların 10'u (%23) LL, 25'i (%57) LS ve 9'u (%20) SS genotipinde bulunmuştur. Allelik dağılımlarında ise L alleli 45 (%51), S alleli ise 43 (%49) olarak bulunmuştur. SLC6A4 allellerinin dağılımı nöropsikiyatrik bozukluklar gösteren bireylerde ve sağlıklı bireylerde farklılık gösterdiği bildirilmiştir. Bu çalışma LL genotip ve L allelinin Türk profesyonel futbolcularda SS genotip ve S alleline oranla daha sık olduğunu gösteren ilk araştırma niteliğindedir.
