We treated 10 patients with histologically confirmed osteosarcoma with three courses of intraarterial administration of CDDP as preoperative chemotherapy, in dose of 100 mg/m2 every three weeks. Out of the 10 cases, 6 cases showed decrease in size of soft part tumor, and 8 cases showed significant decrease of serum Al-p. On X-ray and CT scan examination, complete marginal sclerosis was recognized in 2 cases, and incomplete marginal sclerosis in 6 cases. On analysis of the histological mapping,
more » ... cases showed excellent (necrotic area over 95%), 5 cases good (between 50% and 95%), and 2 cases poor (less 50%). Areas of marginal sclerosis on CT scan showed continuous reactive woven bone formation without any viable tumor cells, covered by fibrous connective 'tissue mimicking periosteum. Histologic study of the pulmonary metastatic nodules indicated viable tumor cell proliferation intermingled by tumor osteoid formation. On analysis of cytophotometric study of nuclear DNA content from biopsy specimens, all cases showed hyperploidy and aneuploidy pattern.
