A microsome-dependent binding of benzo[a]pyrene to DNA

Benzo[a] pyrene-3H incubated in the presence of rat liver microsomes with calf thymus DNA binds covalently to the DNA. The reaction requires reduced nicotinamide adenine di nucleotide phosphate and is greater when the microsomes are obtained from rats pretreated with polycycic hydrocarbons. The binding is due to the formation of unknown metabolite(s) which then may bind to the DNA in the absence of microsomes.
