Mode of Action of the Fungicide Guazatine in Alternaria kikuchiana

The effects of an agricultural fungicide, guazatine triacetate [1, l'-iminiodi(octamethylene)diguanidinium triacetate, BEFRAN(R)] on the biosynthesis of protein, DNA, RNA, cell wall and lipid, and on the respiration of cells were investigated in Alternaria kikuchiana. The incorporations of [U-14C]amino acid mixture, [2-14C]thymidine, [2-14C]uridine and [l-14C]Dglucosamine into protein, DNA, RNA and chitin respectively, were not significantly affected in mycelial cells of A. kikuchiana, by the
more » ... eatment of 10ppm guazatine triacetate, of which the concentration was ID50 value for mycelial growth. The incorporation of [U-14C]D-glucose into cell wall was only moderately inhibited by 10ppm guazatine triacetate after an incubation of 90 min. A similar result was obtained in the case of incorporation of [l-14C]sodium acetate into cell wall. On the other hand, the incorporations of [U-14C]D-glucose, [l-14C]sodium acetate and [methyl-14C]methionine into total lipid were remarkably inhibited by 10 ppm guazatine triacetate. The inhibitions of incorporation were found in the simple lipid fraction after the separative analysis of both simple and complex lipids. Oxygen uptake by the intact cells of A. kikuchiana was hardly inhibited by the fungicide. These data suggest that the primary antifungal action of guazatine triacetate is the inhibition of lipid biosynthesis in A. kikuchiana.
