EVALUATION OF GENOMIC DNA SPIN-50 METHOD FOR AMPLIFICATION OF ITS2 GENE IN SPIDERS

Though various ecological studies have been done in the past on spiders, still the establishment of genetic variations and their evolutionary significance are unknown to a larger extent. Thus analysing genetic variations using DNA polymorphisms has become an important tool. In this study we have compared the efficiency of two widely used DNA extraction methods Guanidium Iso thiocyanate (GITC) and Cetyl tri methyl ammonium bromide (CTAB) with the Chromous Genomic DNA Spin-50 isolation method for
more » ... rapid purification of DNA. Efficiency of the extraction was compared in terms of quantity and quality of DNA obtained for ribosomal internal transcribed spacer (ITS2) marker analysis. Our analysis showed a negligible difference with respect to quantity of DNA extracted and significant difference with regard to the amplification and reproducibility of PCR products, DNA extracted from the Genomic DNA Spin-50 method was found to be more economical compared to home spun protocols.
