Quantification of Histamine by Postcolumn Fluorescence Detection High-Performance Liquid Chromatography Using Orthophthalaldehyde in Tetrahydrofuran and Reaction Mechanism

For quantification of histamine, we developed a highly specific and sensitive method of postcolumn fluorescence detection HPLC using a tetrahydrofuran solution of orthophthalaldehyde. Its detection limit was 2 -5 pg. This method was applied to the measurement of histamine in human blood and urine samples. Analysis by LC/MS and NMR suggested that the fluorophore in this method has an isoindole structure.
