Determination of Knowledge of Caregivers about Dietary Practices of Autistic Children

and Objective: Autism belongs to a complex neurodevelopmental disorder. The lack of knowledge about dietary practices of autistic caregivers is perceived as the triggering factor of 'Autistic Spectrum Disorder (ASD). In this particular 'Knowledge, Attitude and Practice (KAP) Survey' the knowledge (K) of nutrition, attitude (A) of children towards such nutrition and dietary practices (P) among the caregivers of autistic children. more » ... he knowledge questionnaire was designed to facilitate the caregivers to determine their knowledge regarding the dietary patterns of autistic children. Methods: This quasi-experimental study comprised of 50 caregivers (particularly mothers, and fathers in a few instances) of autistic children enrolled at Amin Maktab Center for Special Education Lahore. The nutrition education program was developed and participants were enrolled. Analysis of the change in knowledge, attitude and practice of autistic caregivers through pre-testing and post-testing at a gap of four months was carried out using statistical tests. Results: The pre-testing and post-testing phase of the study revealed significant improvement in (a) basic knowledge about nutritional needs of autistic children (22% vs. 100%), (b) practicing patient-tailored special diet (34% vs. 92%) and (c) resolution of food allergies (62% vs. 16%) in autistic children. The most important benchmark in this regard was Body Mass Index (BMI) of children where deranged BMI improved significantly (p = 0.000) to normal BMI in pre-and post-testing phases. Conclusions: Lack of knowledge is higher among aforementioned caregivers of concerned autistic children. Adoption of nutrition education programs by anutistic caregivers helps in creating optimal knowledge, attitude and practices regarding nutritional needs of autistic children for effective targeted behavior and management.

Mutational analysis on predicting the impact of high-risk SNPs in human secretary phospholipase A2 receptor (PLA2R1)

PLA2R1 is a transmembrane glycoprotein that acts as an endogenous ligand which stimulates the processes including cell proliferation and cell migration. The SNPs in PLA2R1 is associated with idiopathic membranous nephropathy which is an autoimmune kidney disorder. The present study aimed to explore the structure-function analysis of high risk SNPs in PLA2R1 by using 12 different computational tools. more » ... er subjected to evolutionary conservation analysis. Those SNPs which were predicted as deleterious in both categories were further considered for structure based analysis. The resultant SNPs were C1096S, C545S, C664S, F1257L, F734S, I1174T, I1114T, P177S, P384S, W1198G, W1328G, W692C, W692L, W962R, Y499H. One functional domain of PLA2R1 is already modelled in PDB (6JLI), the full 3D structure of the protein was predicted using I-TASSER homology modelling tool. The stability analysis, structure superimposition, RMSD calculation and docking studies were carried out. The structural analysis predicted four mutations F734S, F1246L, I1174T, W1198G as damaging to the structure of the protein. All these mutations are occurring at the conserved region of CTL domain hence are more likely to abolish the function of the protein. Up to the best of our knowledge, this is the first study that provides in-depth and in-silico analysis of deleterious mutations on structure and function of PLA2R1.

Calmodulin-dependent protein kinase kinase 2 (CAMKK2) is a protein kinase that belongs to the serine/threonine kinase family. more » ... silico bioinformatics analysis that combines sequence- and structure-based predictions to mark a SNP as damaging or neutral. The combined results from sequence-based, evolutionary conservation-based, and consensus-based tools have predicted a total of 18 nsSNPs as deleterious, and these nsSNPs were further subjected to structure-based analysis. The six mutant models of V195A, V249M, R311C, F366Y, P389T, and W445C showed a higher deviation from the wildtype protein model and hence were further taken for docking studies. The molecular docking analysis has predicted that these mutations will also be disruptive to the protein-protein interactions between CAMKK2 and PRKAG1 which will create an evident reduction in the kinase activity. The current study has enlightened us that a few of the significant mutations are prime candidates in CAMKK2 which could be the fundamental cause of various bipolar and psychiatric disorders. This is the first detailed study that predicts the deleterious nsSNPs in CAMKK2 and contributes positively in providing a better understanding of disease mechanisms.

Association of musculoskeletal discomfort with the use of high heeled shoes in females

Objective: To determine the association of musculoskeletal discomfort with the high heeled shoes related variables in females who use such footwear frequently. Methods: A cross sectional study was conducted on 174 female residents of Islamabad and Rawalpindi, for a time period stretching from July 1, 2018 to December 31, 2018. more » ... cale (NPRS) were used as outcome measures to assess the musculoskeletal discomfort and a self-structured questionnaire was used to obtain information related to the high heeled shoes e.g. duration and frequency of high heeled shoe use. Data was analyzed using SPSS version 21. Results: The data analysis showed a positive association of shoulders' discomfort with length of high heeled shoes used by women (p value= 0.03<0.05). Moreover, a positive association was also found between an occurrence of upper back discomfort and length of the heeled shoes (p value= 0.01<0.05). However, no significant association was found between pain intensity and length of the shoe' heel, frequency and duration of high heeled shoe use. Conclusion: The shoulders' and upper back discomfort in females who frequently used high heeled shoes was determined to be associated with the length of the heels in shoes. In Pakistan, due to legal and religious association, cases of attempted suicides are underreported in Pakistan, yet it is essential to have accurate data so that the causality leading to this national tragedy can be studied and minimized. Psychiatric disorders leading to suicide is largely neglected and under-researched in Pakistan. In this cross-sectional survey, patients brought to emergency with attempted suicide were enrolled in the study, after taking informed consent from the attendant. After initial treatment, the patient's clinical history was sought via a General Health Questionnaire-28. Once recognized, participants underwent detailed psychiatric evaluation and mental state examination. Three hundred and fifty-two (352) patients were brought to the emergency with attempted suicide, of which 249 (70.7%) patients were identified with psychiatric morbidity. The most common psychiatric disorders were mood disorder (32.1%), comorbid psychiatric disorder (20.4%), and anxiety disorders (18.4%). Our study also showed that the prevalence of comorbid psychiatric disorders was significantly higher in females as compared to males, whereas substance use disorder was more common in males. The suicide rate has increased globally due to associated psychiatric disorders. Patients inflicting self-harm or failing at suicidal attempt are inclined towards attempting suicide in future. However, the social stigma associated with psychiatric disorders has heavily affected the process of successfully identifying and treating such patients. Along with focused long-term treatment, follow-up, and enhanced surveillance programs, mass awareness campaigns should be conducted to improve the knowledge and outlook of the general population towards psychiatric disorders.

In plants, F-box proteins (FBPs) constitute one of the largest superfamilies of regulatory proteins. Most F-box proteins are shown to be an integral part of SCF complexes, which carry out the degradation of proteins and regulate diverse important biological processes. Anthers and pollen development have a huge importance in crop breeding. Despite the vast diversity of FBPs in Arabidopsis male reproductive organs, their role in anther and pollen development is not much explored. Moreover, a
more » ... ard nomenclature for naming FBPs is also lacking. Here, we propose a standard nomenclature for naming the FBPs of Arabidopsis thaliana uniformly and carry out a systematic analysis of sperm cell-specific FBP gene, i.e., 3p.AtFBP113 due to its reported high and preferential expression, for detailed functional annotation. The results revealed that 3p.AtFBP113 is located on the small arm of chromosome and encodes 397 amino acid long soluble, stable, and hydrophilic protein with the possibility of localization in various cellular compartments. The presence of the C-terminal F-box associated domain (FBA) with immunoglobulin-like fold anticipated its role in protein binding. Gene ontology based functional annotation and tissue-specific gene co-expression analysis further strengthened its role in protein binding and ubiquitination. Moreover, various potential post/co-translational modifications were anticipated and the predicted tertiary structure also showed the presence of characteristic domains and fold. Thus, the outcomes of the study will be useful in developing a better understating of the function of 3p.AtFBP113 during the process of pollen development, which will be helpful for targeting the gene for manipulation of male fertility that has immense importance in hybrid breeding.

Drugs are small molecules that generally work by binding to its target which is often a protein. This ligand molecule binding helps in the treatment of various diseases. Major obstacle to treat complex diseases is the phenomena underlying drug resistance mechanisms which are not fully understood so far. Retrieving information on these processes can be challenging as scientific literature has huge pool of data and extracting the required information has always been a laborious task. We developed an online pipeline ZK DrugResist that automatically extracts PubMed abstracts of drug resistance paired with either mutation or expression for a given disease. Our classifier showed 97.7% accuracy with 93.5% recall and 96.5% F-measure. This system saves plenty of time in terms of data mining and also reduces efforts in retrieving information from online resources.

Cancer is a metabolic disorder and in energy metabolism, pyruvate dehydrogenase kinases (PDKs) play a very vital role, which make them important candidate for involvement in cancer. The bioinformatics analysis performed on ovarian cancer data sets taken from gene expression Omnibus proved the up regulation of PDK2 and PDK4.
