2013 BMC Medical Genetics
The human paraoxonase (PON) gene family has three isoforms: PON1, PON2 and PON3. These genes are implicated as potential risk factors of cerebrovascular disease and can prevent oxidative modification of low-density lipoproteins and atherosclerosis. This study evaluated the association between the genetic variants of all three PON genes and the risks of total stroke, ischemic stroke and hemorrhagic stroke in the Han Chinese population. Methods: A total of 1016 subjects were recruited, including
more » ... 08 healthy controls and 498 patients (328 with ischemic stroke and 170 with hemorrhagic stroke). A total of 11 single nucleotide polymorphisms (SNPs) covering the PON genes were genotyped for statistical analysis. Two of the 11 SNPs (rs662 and rs854560) were contextualized in a meta-analysis of ischemic stroke. Results: The presence of rs705381 (−162) in the promoter region of PON1 was significantly associated with total stroke (P adjusted = 0.0007, OR = 0.57 [95% CI = 0.41-0.79]) and ischemic stroke (P adjusted = 0.0017, OR = 0.54 [95% CI = 0.37-0.79]) when analyzed using a dominant model, but was not associated with hemorrhagic stroke. There was also a nominal association between rs854571 (−824) and total stroke. Meta-analysis demonstrated a significant nominal association between rs662 and ischemic stroke, but there was no evidence of an association between rs662 and ischemic stroke risk in a single site association study. Conclusions: These findings indicate that polymorphisms of PON1 gene may be a risk factor of stroke.

Mechanism of pulse magneto-oscillation grain refinement on pure Al

Pei Ning, Gong Yongyong, Li Renxing
2011 China Foundry
Pulse magneto-oscillation (PMO) was developed as a novel technique to refine the solidification structure of pure aluminium. Its grain refining mechanism was proposed. The PMO refinement mechanism is that the nucleus falls off from the mould wall and drifts into the melt under the action of PMO. The solidification structure of Al melt depends on the linear electric current density, and also the discharge and oscillation frequencies. The radial pressure of PMO sound wave is the major factor that

2022 Molecular Brain
AbstractEpilepsy has been extensively studied as a common neurological disease. Efforts have been made on rodent and other animal models to reveal the pathogenic mechanisms of epilepsy and develop new drugs for treatment. However, the features of current epilepsy models cannot fully mimic different types of epilepsy in humans, hence non-human primate models of epilepsy are required. The common marmoset (Callithrix jacchus) is a New World monkey that is widely used to study brain function. Here,
more » ... we present a natural marmoset model of generalized epilepsy. In this unique marmoset family, generalized epilepsy was successfully induced by handling operations in some individuals. We mapped the marmoset family with handling-sensitive epilepsy and found that the epileptic phenotype can be inherited. These marmosets were more sensitive to the epilepsy inducers pentylenetetrazol. Using electrocorticogram (ECoG) recordings, we detected epileptiform discharge in marmosets with a history of seizures. In summary, we report a family of marmosets with generalized seizures induced by handling operations. This epileptic marmoset family provides insights to better understand the mechanism of generalized epilepsy and helps to develop new therapeutic methods.

2019 Materials Research
Microstructural evolution, interfacial reaction and fracture during vacuum brazing of 50Mo-50Re alloys using amorphous alloy Fe-5.6Si-2.6B(wt%) were investigated. A binary intermetallic compound, σ(Mo 2 Re 3 ) formed as parallel contiguous layers in the diffusion zone. In addition, Fe from the braze alloy, in combination with dissolved Mo from the substrate, formed two layers of binary compounds μ-Mo 6 Fe 7 and λ-MoFe 2 , adjacent to diffusion zone. The unreacted Fe-based filler metal

2020 BMC Psychiatry
Recent genome-wide association study showed rs10940346 locus near EMB gene was significantly associated with schizophrenia and suggested that EMB gene is one of the potentially causal genes for schizophrenia, but no causal variant has been identified. Our study aims to further verify EMB gene is a susceptibility gene for schizophrenia and to identify potentially causal variants in EMB gene that lead to schizophrenia. Targeted sequencing for the un-translated region and all exons of EMB gene was
more » ... performed among 1803 patients with schizophrenia and 997 healthy controls recruited from Chinese Han population. A total of 58 high-quality variants were identified in case and control groups. Seven of them are nonsynonymous rare variations, EMB: p.(Ala52Thr), p.(Glu66Gly), p.(Ser93Cys), p.(Ala118Val), p.(Ile131Met), p.(Gly163Arg) and p.(Arg238Tyr), but none of them reached statistical significance. Among them, p.(Ile131Met), p.(Gly163Arg) and p.(Arg238Tyr), were predicted to be deleterious variants. In addition, a common variant, rs3933097 located in 3'-UTR of EMB gene, achieved allelic and genotypic significance with schizophrenia (Pallele = 3.82 × 10- 6, Pgenotype = 3.18 × 10- 5). Our research first presented a comprehensive mutation spectrum of exons and un-translated region in EMB gene for schizophrenia and provided additional evidence of EMB gene being a susceptibility gene for schizophrenia. However, further functional validations are necessary to reveal its role in the etiology of schizophrenia.

Engineering of a Pluronic F127 functionalized magnetite/graphene nanohybrid for chemophototherapy

Yongyong Li, Jiaqiang Liu, Haiqing Dong, Guangzhen Liu, Haiqing Hu
2014 Nanotechnology
In this study, a multifunctional graphene based nanohybrid, termed as GN/Fe 3 O 4 /PF127, is engineered via a facile one-pot process consisting of simultaneous reduction of graphene oxide/Fe 3 O 4 and subsequent assembly of Pluronic F127 (PF127) onto graphene nanosheets (GNs). The unique aromatic and planar structure of GNs allows the attachment of multiple functional components including MRI contrast agent (Fe 3 O 4 nanoparticles) and an aromatic anticancer drug like doxorubicin (DOX), as well
more » ... as PF127 coating which imparts physiological dispersivity and stability to the nanohybrid. The successful assembly process is revealed by TEM observation, size and FITR monitoring. In contrast with the primitive graphene or its oxide derivative, the resulting GN/Fe 3 O 4 /PF127 nanohybrids have shown high biological dispersion and MRI effect for diagnosis due to the incorporation of superparamagnetic Fe 3 O 4 nanoparticles without evident cytotoxicity. Moreover, the GN/Fe 3 O 4 /PF127 nanohybrid exhibits a photothermal effect due to the considerable optical absorption in the near-infrared region of GNs. The GN/Fe 3 O 4 /PF127 nanohybrid could be a further platform for chemophototherapy assisted by the therapeutic DOX. Cellular toxicity assays indicated that the DOX-loaded GN/Fe 3 O 4 /PF127 nanohybrid showed a remarkable cytotoxicity to HeLa cells and the cytotoxic effect was intensified when subjected to photoirradiation. Confocal laser scanning microscopy (CLSM) and flow cytometric analysis (FCAS) revealed that the nanohybrid could be easily uptaken into HeLa cells.
