Forest aboveground biomass estimation using polarization coherence tomography and PolSAR segmentation

Forest aboveground biomass (AGB) is essential for monitoring the carbon cycle budget and climate change. This study proposes a method for the estimation of forest AGB based on polarization coherence tomography (PCT) and polarimetric synthetic aperture radar (PolSAR) segmentation. The data used are the single-baseline polarimetric SAR interferometry data acquired by the German Aerospace Center's E-SAR sensor at the Lband over the city of Traunstein and its vicinity in 2003. First, vertical
more &raquo; ... ure tomographic profiles (relative reflectivity distribution in vertical direction) were produced by PCT for each pixel and then averaged within each field-surveyed forest stand to obtain the mean tomographic profile. Next, the mean vertical tomographic profiles were parameterized by 10 shape parameters. Several models for biomass estimation were designed based on the relationships between the definition parameters and the in situ measurements using backward step-wise regression and the M-fold (10-fold) crossvalidation method. The best model was chosen by evaluation criteria such as R, root mean square error (RMSE), R 2 , etc. Forest polygons (objects) were produced by image segmentation using forest heights, A 1 (produced by coherence optimization representing double-bounce scattering mechanism), A 2 (produced by coherence optimization representing the volume scattering mechanism), and the volume scattering mechanism class (produced by Freeman-Durden decomposition and Whishart classification). Finally, the selected model was used to estimate the AGB of each forest polygon and was validated using ground-measured biomass with R 2 of 0.883 and RMSE of 39.98 tons ha −1 . The results show that the proposed method works well for the estimation of forest AGB. No saturation phenomena have been observed even for the forest stands with AGB larger than 500 tons ha −1 .

Effect of Sirtuin-1 on Synaptic Plasticity in Nucleus Accumbens in a Rat Model of Heroin Addiction

Synaptic plasticity plays an important role in the process of addiction. This study investigated the relationship between synaptic plasticity and changes in addictive behavior and examined the expression of synaptic plasticity-associated proteins and genes in the nucleus accumbens (NAc) region in different rat models. more &raquo; ... ern blotting, and transmission electron microscopy were used to detect changes in synaptic plasticity-related gene and protein expression, and changes in the ultrastructure of synapses, in the NAc. Results: Naloxone withdrawal symptoms appeared in the SIRT1-overexpression group. In the SIRT1-silenced group the symptoms were reduced. Immunohistochemistry and Western blotting results showed that FOXO1 expression decreased in the heroin addiction (HA) group but increased in the SIRT1-silenced group (p<0.05). The expression of Cdk5, Nf-kB, PSD95, and Syn was enhanced in the HA group (p<0.05) and further increased in the SIRT1-overexpression group but were reduced in the SIRT1-silenced group (p<0.05). The number of synapses increased in the HA group (p<0.05) along with mitochondrial swelling in the presynaptic membrane and obscuring of the synaptic cleft. Conclusions: SIRT1 and other synaptic plasticity-related genes in NAc are involved in the regulation of heroin addiction. SIRT1 overexpression can increase behavioral sensitization in the NAc of rats, and SIRT1 silencing might ease withdrawal symptoms and reduce conditioned place preferences.

Decreased fructose-1,6-bisphosphatase-2 expression promotes glycolysis and growth in gastric cancer cells

Increasing evidence suggests that cancer is a metabolic disease. Here, we investigated the potential role of fructose-1,6-bisphosphatase-2 (FBP2), the enzyme that catalyses the hydrolysis of fructose-1,6-bisphosphate to fructose-6-phosphate and inorganic phosphate in glucose metabolism, in gastric cancer (GC) development. Results: Our data indicated that FBP2 was downregulated in GC tissues (86.2%, 100/116), and absent or low FBP2 expression in GC tissues was correlated with poor survival of GC
more &raquo; ... patients (P = 0.019). Conversely, ectopic expression of FBP2 in GC cells activated AMP-activated protein kinase (AMPK) signalling, inhibited the Akt-mTOR pathway, suppressed glucose metabolism, enhanced apoptosis, and reduced cell proliferation. Bisulphite genomic sequencing (BGS) in gastric cancer cell lines revealed that the FBP2 promoter region was densely methylated, and treatment of GC cells with the demethylation reagent, 5-aza-2-deoxycytidine (5-Aza), led to an increase in FBP2 expression. Importantly, forced expression of FBP2 abrogated tumour formation of these GC cells in nude mice. Conclusion: Our results indicate that FBP2 does negatively regulate cell growth, and reduced expression of FBP2 may contribute to carcinogenesis for GC. These findings suggest that restoration of FBP2 expression can be a promising strategy for the target therapy of GC.

more &raquo; ... sion profiles of GSE28100 [the Gene Expression Omnibus (GEO) accession number] were downloaded from the GEO database and an independent sample t-test was used to identify statistical differences between the DE-miRNAs of the oral cancer patients and the healthy control subjects. The target genes of DE-miRNA were retrieved from the miRecords database. Furthermore, a protein-protein interaction network was constructed using the Search Tools for the Retrieval of Interacting Genes database and Cytoscape software. A total of 15 DE-miRNAs were identified and among them, hsa-miR-15a drew specific attention. Gene Ontology analysis revealed that the target genes of fibroblast growth factor (FGF)2 are involved in the progression of oral cancer. Furthermore, functional analysis indicated that the FGF-receptor signaling pathway was significantly upregulated in oral cancer. hsa-miR-15a is important in the regulation of oral cancer and thus, may present a potential biomarker for the prediction of oral cancer progression.

Comparison of dry and wet deposition of particulate matter in near-surface waters during summer

Atmospheric particulate matter (PM) deposition which involves both dry and wet processes is an important means of controlling air pollution. To investigate the characteristics of dry and wet deposition in wetlands, PM concentrations and meteorological conditions were monitored during summer at heights of 1.5 m, 6 m and 10 m above ground level at Cuihu Wetland (Beijing, China) in order to assess the efficiency of PM2.5 (particles with an aerodynamic size of <2.5 μm) and PM10 (particles with an
more &raquo; ... rodynamic size of <10 μm) removal. The results showed: Daily concentrations of PM, dry deposition velocities and fluxes changed with the same variation trend. The daily average deposition velocity for PM10 (3.19 ± 1.18 cmÁs -1 ) was almost 10 times that of PM2.5 (0.32 ± 0.33 cmÁs -1 ). For PM2.5, the following dry deposition fluxes were recorded: 10 m (0.170 ± 0.463 μgÁm -2 Ás -1 ) > 6 m (0.007 ± 0.003 μgÁm -2 Ás -1 ) > 1.5 m (0.005 ± 0.002 μgÁm -2 Ás -1 ). And the following deposition fluxes for PM10 were recorded: 10 m (2.163 ± 2.941 μgÁm -2 Ás -1 ) > 1.5 m (1.565 ± 0.872 μgÁm -2 Ás -1 ) > 6 m (0.987 ± 0.595 μgÁm -2 Ás -1 ). In the case of wet deposition, the relative deposition fluxes for PM2.5 and PM10 were 1.5 m > 10 m > 6 m, i.e. there was very little difference between the fluxes for PM2.5 (0.688 ± 0.069 μgÁm -2 Ás -1 ) and for PM10 (0.904 ± 0.103 μgÁm -2 Ás -1 ). It was also noted that rainfall intensity and PM diameter influenced wet deposition efficiency. Dry deposition (63%) was more tilted towards removing PM10 than was the case for wet deposition (37%). Synthesis and biological evaluation of amino acid derivatives containing chrysin that induce apoptosis

A series of chrysin amino acid derivatives were synthesized to evaluate for their antiproliferative activities against several cancer cell lines. Among the compounds tested, N-(2-((5-hydroxy-4-oxo-2-phenyl-4H-chromen-7-yl)oxy)octanoyl)-L-leucine methyl ester(5d) presented a good anti-proliferative activity in MDA-MB-231 and MCF-7 cells. Flow cytometry analysis showed that 5d induced apoptosis and prolonged cell cycle progression in MDA-MB-231 and MCF-7 cells. Western blot analysis showed that
more &raquo; ... significantly inhibited Akt phosphorylation (Ser473) in MDA-MB-231 and MCF-7 cells. In addition, 5d treatment markedly downregulated Bcl-2 and upregulated Bax in a dose-dependent manner. In vitro caspase activation assay showed that 5d induced apoptosis of MDA-MB-231 cells by enhancing caspase 3/7 activity. The regulatory effect of 5d on apoptosis of MDA-MB-231 and MCF-7 cells may be induced by mitochondrial apoptosis pathway. This study is of great significance for designing and developing more effective chrysin amino acid derivatives.

MiR-802 alleviates lipopolysaccharide-induced acute lung injury by targeting Peli2

Acute respiratory distress syndrome (ARDS) is a life-threatening medical condition. It is characterized by serious lung inflammation or injury. Characterizing novel miRNAs implicated in ARDS pathogenesis may provide new therapeutic strategy for managing ARDS. We employed LPS-induced lung injury model to profile miRNAs associated with ARDS. We isolated one miRNA candidate and characterized its role in lipopolysaccharide (LPS)-induced proinflammatory cytokine production in lung macrophages. We
more &raquo; ... ther evaluated its functional role in ARDS model by assessing histological change, neutrophil activation, tissue permeability and tumor necrosis factor alpha (TNFα) production. We also characterized its downstream target using luciferase assay, Western blotting, enzyme-linked immunosorbent assay and cell inflammation assay. Microarray profiling revealed miR-802 was significantly downregulated in ARDS mouse model. LPS-induced miR-802 downregulation was confirmed in lung macrophages. Overexpression of miR-802 significantly suppressed LPS-induced inflammatory cytokine production in vitro and alleviates LPS-induced acute lung injury in vivo. Peli2 was identified as a downstream target of miR-802 and found upregulated in ARDS model. Overexpressing Peli2 abolished the antagonizing effect of miR-802 on LPS-mediated inflammatory response. MiR-802 carried a protective role against LPS-induced acute lung injury by downregulating Peli2. MiR-802/Peli2 axis may act as intervening targets to manage ARDS.
