Digital Data Preservation for Scholarly Publications in Astronomy
2008
International Journal of Digital Curation
Astronomy is similar to other scientific disciplines in that scholarly publication relies on the presentation and interpretation of data. But although astronomy now has archives for its primary research telescopes and associated surveys, the highly processed data that is presented in the peer-reviewed journals and is the basis for final analysis and interpretation is generally not archived and has no permanent repository. We have initiated a project whose goal is to implement an end-to-end

... type system which, through a partnership of a professional society, that society's scholarly publications/publishers, research libraries, and an information technology substrate provided by the Virtual Observatory, will capture high-level digital data as part of the publication process and establish a distributed network of curated, permanent data repositories. The data in this network will be accessible through the research journals, astronomy data centers, and Virtual Observatory data discovery portals.
Toll-Like Receptor Stimulation Induces Higher TNF-α Secretion in Peripheral Blood Mononuclear Cells from Patients with Hyper IgE Syndrome
2008
International Archives of Allergy and Immunology
Hyper IgE syndromes (HIES) are primary immunodeficiency disorders of unknown pathogenesis. Patients are typically affected with 'cold' abscesses of the skin, recurrent cyst-forming pneumonia, chronic mucocutaneous candidiasis and other less frequent features such as progressive skeletal abnormalities. Defective signaling in the Toll-like receptor (TLR) pathways has been suggested as a responsible pathologic mechanism, however, in previous reports, 10 patients revealed no defect in inflammatory

... ytokine responses to different TLR ligands. Here, we report the increase in pro-inflammatory cytokines TNF-␣ and IL-8, following TLR2 and TLR4 stimulation in a larger cohort of 25 additional patients with HIES, and provide a meta-analysis of the TLR data in HIES.
Mutations in STAT3 and diagnostic guidelines for hyper-IgE syndrome
2010
Journal of Allergy and Clinical Immunology
The hyper-IgE syndrome (HIES) is a primary immunodeficiency characterized by infections of the lung and skin, elevated serum IgE, and involvement of the soft and bony tissues. Recently, HIES has been associated with heterozygous dominantnegative mutations in the signal transducer and activator of transcription 3 (STAT3) and severe reductions of T H 17 cells. Objective: To determine whether there is a correlation between the genotype and the phenotype of patients with HIES and to establish

... stic criteria to distinguish between STAT3 mutated and STAT3 wild-type patients. Methods: We collected clinical data, determined T H 17 cell numbers, and sequenced STAT3 in 100 patients with a strong clinical suspicion of HIES and serum IgE >1000 IU/mL. We explored diagnostic criteria by using a machine-learning approach to identify which features best predict a STAT3 mutation. Results: In 64 patients, we identified 31 different STAT3 mutations, 18 of which were novel. These included mutations at splice sites and outside the previously implicated DNA-binding and Src homology 2 domains. A combination of 5 clinical features predicted STAT3 mutations with 85% accuracy. T H 17 cells were profoundly reduced in patients harboring STAT3 mutations, whereas 10 of 13 patients without mutations had low (<1%) T H 17 cells but were distinct by markedly reduced IFN-g-producing CD4 1 T cells. Conclusion: We propose the following diagnostic guidelines for STAT3-deficient HIES. Possible: IgE >1000IU/mL plus a weighted score of clinical features >30 based on recurrent pneumonia, newborn rash, pathologic bone fractures, characteristic face, and high palate. Probable: These characteristics plus lack of T H 17 cells or a family history for definitive HIES. Definitive: These characteristics plus a dominant-negative heterozygous mutation in STAT3. Oncogenic JAK2 V617F causes PD-L1 expression, mediating immune escape in myeloproliferative neoplasms
2018
Science Translational Medicine
Recent evidence has revealed that oncogenic mutations may confer immune escape. A better understanding of how an oncogenic mutation affects immunosuppressive programmed death ligand 1 (PD-L1) expression may help in developing new therapeutic strategies. We show that oncogenic JAK2 (Janus kinase 2) activity caused STAT3 (signal transducer and activator of transcription 3) and STAT5 phosphorylation, which enhanced PD-L1 promoter activity and PD-L1 protein expression in JAK2 V617F -mutant cells,

... ereas blockade of JAK2 reduced PD-L1 expression in myeloid JAK2 V617F -mutant cells. PD-L1 expression was higher on primary cells isolated from patients with JAK2 V617F -myeloproliferative neoplasms (MPNs) compared to healthy individuals and declined upon JAK2 inhibition. JAK2 V617F mutational burden, pSTAT3, and PD-L1 expression were highest in primary MPN patient-derived monocytes, megakaryocytes, and platelets. PD-1 (programmed death receptor 1) inhibition prolonged survival in human MPN xenograft and primary murine MPN models. This effect was dependent on T cells. Mechanistically, PD-L1 surface expression in JAK2 V617F -mutant cells affected metabolism and cell cycle progression of T cells. In summary, we report that in MPN, constitutive JAK2/STAT3/STAT5 activation, mainly in monocytes, megakaryocytes, and platelets, caused PD-L1-mediated immune escape by reducing T cell activation, metabolic activity, and cell cycle progression. The susceptibility of JAK2 V617F -mutant MPN to PD-1 targeting paves the way for immunomodulatory approaches relying on PD-1 inhibition.
