Chemical Constituents from the Barks of Swietenia macrophylla

A new tirucallane-type triterpenoid, swietentol (1), was isolated from the barks of Swietenia macrophylla, together with four known triterpenoids (2-5), five known limonoids (6-10), and other six compounds (11-16). The structure of swietentol (1) was elucidated by extenstive 2D NMR and MS analysis.

Clinical metagenomics assessments improve diagnosis and outcomes in community-acquired pneumonia

Background Identifying the causes of community-acquired pneumonia (CAP) is challenging due to the disease's complex etiology and the limitations of traditional microbiological diagnostic methods. Recent advances in next generation sequencing (NGS)-based metagenomics allow pan-pathogen detection in a single assay, and may have significant advantages over culture-based techniques. Results We conducted a cohort study of 159 CAP patients to assess the diagnostic performance of a clinical

When compared to other techniques, clinical metagenomics detected more pathogens in more CAP cases, and identified a substantial number of polymicrobial infections. Moreover, metagenomics results led to changes in or confirmation of clinical management in 35 of 59 cases; these 35 cases also had significantly improved patient outcomes. Conclusions Clinical metagenomics could be a valuable tool for the diagnosis and treatment of CAP. Complete Genome Sequences of an Avian Orthoreovirus Isolated from Guangxi, China

We report the complete genomic sequences of an avian orthoreovirus, strain GuangxiR1, isolated from a chicken flock in Guangxi Province, southern China, in 2000. Phylogenetic analyses suggest that the strain is closely related to the S1133 strain, which is associated with tenosynovitis, but is far different from strain AVS-B, which is associated with runting-stunting syndrome in broilers.

Monocular Road Planar Parallax Estimation [article]

Estimating the 3D structure of the drivable surface and surrounding environment is a crucial task for assisted and autonomous driving. It is commonly solved either by using expensive 3D sensors such as LiDAR or directly predicting the depth of points via deep learning. Instead of following existing methodologies, we propose Road Planar Parallax Attention Network (RPANet), a new deep neural network for 3D sensing from monocular image sequences based on planar parallax, which takes full advantage
more &raquo; ... of the commonly seen road plane geometry in driving scenes. RPANet takes a pair of images aligned by the homography of the road plane as input and outputs a γ map for 3D reconstruction. Beyond estimating the depth or height, the γ map has a potential to construct a two-dimensional transformation between two consecutive frames while can be easily derived to depth or height. By warping the consecutive frames using the road plane as a reference, the 3D structure can be estimated from the planar parallax and the residual image displacements. Furthermore, to make the network better perceive the displacements caused by planar parallax, we introduce a novel cross-attention module. We sample data from the Waymo Open Dataset and construct data related to planar parallax. Comprehensive experiments are conducted on the sampled dataset to demonstrate the 3D reconstruction accuracy of our approach in challenging scenarios.

Micromechanics-Based Low Cycle Fatigue Life Prediction Model of ECAPed Aluminum Alloy

Ultrafine-grained aluminum alloys (UFG AA) show great potential in the design of fatigue-resistant lightweight alloys, and the methodology to assess low-cycle fatigue (LCF) life remains to be studied. In this work, a micromechanics-based LCF life prediction model is presented by conducting crystal plasticity finite element simulation (CPFEM). The fatigue indicator parameter (FIP) of maximum accumulated equivalent plastic strain energy, modulated by triaxiality, is developed to assess the
more &raquo; ... l damage in the microstructure. Particularly, a new multiaxial strain parameter is proposed by considering the combined influence of the mean strain and non-proportional cyclic additional hardening effect, and then directly embedding into the cyclic J-integral. Finally, the reformulated Manson-Coffin relationship is theoretically constructed by correlating the crack tip opening displacement to the crack propagation equation. The results show the scatter fatigue life of UFG AA6061 is not only related to the inhomogeneous evolution of plastic deformation but also to the local stress state. Since the proposed approach considers both the deformation mechanisms at the micro-scale and the corresponding macroscopic responses, it can predict the LCF life of UFG AA with reasonable accuracy.

A new shock-capturing scheme for stiff detonation waves problems [article]

A new approach to prevent spurious behavior caused by conventional shock-capturing schemes when solving stiff detonation waves problems is introduced in the present work. Due to smearing of discontinuous solution by the excessive numerical dissipation, conventional shock-capturing schemes have difficulties to obtain the correct location of detonation front without enough grids resolution. To overcome the excessive numerical errors around discontinuities by traditional discretized schemes used
more &raquo; ... non-reacting high speed compressible flow, we introduce a new shock-capturing scheme in which besides linear function constructed by MUSCL (Monotone Upstream-centered Schemes for Conservation Law) scheme, a step like THINC (Tangent of Hyperbola for INterface Capturing) function is also employed as another candidate in the reconstruction process. The final reconstruction function is determined by boundary variation diminishing (BVD) algorithm by which numerical dissipation around discontinuities can be reduced significantly. The new resulted shock-capturing scheme is named MUSCL-THINC-BVD. One- and two-dimensional comparative numerical tests about stiff detonation waves problems are conducted with the 5th order WENO (Weighted Essentially Non-Oscillatory) and MUSCL-THINC-BVD scheme respectively, which show MUSCL-THINC-BVD scheme can capture the correct position of detonation waves with improved resolution while WENO scheme, in spite of higher order, produces spurious waves. Compared with other existing methods which involves extra treatments by accepting the smeared out discontinuities profiles, the current method obtain the correct but also sharp detonation front by fundamentally reducing numerical dissipation errors from shock-capturing schemes. Thus the proposed approach is an effective but simple method to solve stiff detonation problems.

Major depressive disorder is a serious and complex mental illness, and multiple brain regions are involved in its pathogenesis. There is increasing evidence that the amygdala is involved in depression; however, the underlying molecular mechanisms remain unclear. Differential analysis identified 42 metabolites and 171 proteins that were differentially expressed in the CUMS and control groups. Integrated analyses revealed two major changes in the amygdala of CUMS rats: (1) perturbations in amino acids and carbohydrate metabolism, transport-/catabolism-related proteins activity, and metabolic enzyme activity; (2) abnormal expression of synaptogenesis and oxidative phosphorylation-associated proteins.

Dynamic forecasting of Zika epidemics using Google Trends [article]

We developed a dynamic forecasting model for Zika virus (ZIKV), based on real-time online search data from Google Trends (GTs). It was designed to provide Zika virus disease (ZVD) surveillance for Health Departments with early warning, and predictions of numbers of infection cases, which would allow them sufficient time to implement interventions. We used correlation data from ZIKV epidemics and Zika-related online search in GTs between 12 February and 25 August 2016 to construct an
more &raquo; ... ve integrated moving average (ARIMA) model (0, 1, 3) for the dynamic estimation of ZIKV outbreaks. The online search data acted as an external regressor in the forecasting model, and was used with the historical ZVD epidemic data to improve the quality of the predictions of disease outbreaks. Our results showed a strong correlation between Zika-related GTs and the cumulative numbers of reported cases, both confirmed and suspected (both p<0.001; Pearson Product-Moment Correlation analysis). The predictive cumulative numbers of confirmed and suspected cases increased steadily to reach 148,510 (95% CI: 126,826-170,195) and 602,721 (95% CI: 582,753-622,689), respectively, in 21 October 2016. Integer-valued autoregression provides a useful base predictive model for ZVD cases. This is enhanced by the incorporation of GTs data, confirming the prognostic utility of search query based surveillance. This accessible and flexible dynamic forecast model could be used in the monitoring of ZVD to provide advanced warning of future ZIKV outbreaks.

The axisymmetric multiple-relaxation-time lattice Boltzmann (LB) method is used to study the acoustic levitation of a rigid disk sample in a closed cylindrical resonant chamber. The simulation results show that the resonant cavity length L is equal to 0.499λ for (001) mode, and the resonance shift δL is approximately equal to −0.9 with a disk sample located in the chamber center, which accord with the analytical results derived from linear acoustics. The LB method naturally includes the
more &raquo; ... y and resonance shift during the simulation of acoustic levitation force on the disk sample, which gives the results not only consistent with the theory in magnitude, but also coherent with the experiments in more details. Some of the nonlinear effects associated with acoustic levitation, such as waveform distortion, acoustic streaming, and radiation pressure, are also revealed by the LB simulation.

Targeted Co-Delivery of siRNA and Methotrexate for Tumor Therapy via Mixed Micelles

A combination of chemotherapeutic drugs and siRNA is emerging as a new modality for cancer therapy. A safe and effective carrier platform is needed for combination drug delivery. Here, a functionalized mixed micelle-based delivery system was developed for targeted co-delivery of methotrexate (MTX) and survivin siRNA. Linolenic acid (LA) was separately conjugated to branched polyethlenimine (b-PEI) and methoxy-polyethyleneglycol (mPEG). MTX was then conjugated to LA-modified b-PEI (MTX-bPEI-LA)
more &raquo; ... o form a functionalized polymer-drug conjugate. Functionalized mixed micelles (M-MTX) were obtained by the self-assembly of MTX-bPEI-LA and LA-modified mPEG (mPEG-LA). M-MTX had a narrow particle size distribution and could successfully condense siRNA at an N/P ratio of 16/1. M-MTX/siRNA was selectively taken up by HeLa cells overexpressing the folate receptor (FR) and facilitated the release of the siRNA into the cytoplasm. In vitro, M-MTX/siRNA produced a synergy between MTX and survivin siRNA and markedly suppressed survivin protein expression. In tumor-bearing mice, M-MTX/Cy5-siRNA showed an elevated tumor uptake. In addition, M-MTX/siRNA inhibited tumor growth. Immunohistochemistry and a western blot analysis showed a significant target gene downregulation. In conclusion, M-MTX/siRNA was highly effective as a delivery system and may serve as a model for the targeted co-delivery of therapeutic agents.

Effect of Preheating Temperature on Geometry and Mechanical Properties of Laser Cladding-Based Stellite 6/WC Coating

The effect of 60Si2Mn substrate preheating on the forming quality and mechanical properties of cobalt-based tungsten carbide composite coating was investigated. Substrate preheating was divided into four classes (room temperature, 150 °C, 250 °C, and 350 °C). The morphology, microstructure, and distribution of elements of the coating were analyzed using a two-color laser handheld 3D scanner, a scanning electron microscope (SEM), and an energy dispersive X-ray spectrometer (EDX), respectively.
more &raquo; ... e hardness and wear properties of the cladding layer were characterized through a microhardness tester and a friction wear experiment. The research results show that the substrate preheating temperature is directly proportional to the height of the composite coating. The solidification characteristics of the Stellite 6/WC cladding layer structure are not obviously changed at substrate preheating temperatures of room temperature, 150 °C, and 250 °C. The solidified structure is even more complex at a substrate preheating temperature of 350 °C. At this moment, the microstructure of the cladding layer is mainly various blocky, petaloid, and flower-like precipitates. The hardness and wear properties of the cladding layer are optimal at a substrate preheating temperature of 350 °C in terms of mechanical properties.
