Difficult management of severe Medication-Related Osteonecrosis of the Jaws in a denosumab treated osteoporotic patient, a case report

In a patient treated with denosumab for osteoporosis, a case of stage II MRONJ occurred after 3 months from implant placement and subsequent implant loss. The first surgical approach led to a worsening at Stage III. A second surgery with a conservative approach was performed more than 6 months after the last denosumab injection obtaining therefore the definitive healing. Pharmacokinetics and surgical approach should be considered in MRONJ treatment scheduling.

Salt stress in popcorn genotypes trigger changes of antioxidant enzymes

Salinity is a major problem in agriculture because it can alter the metabolism of plants and affect crop yield. This study aimed to evaluate the effect of NaCl on growth, key antioxidants and changes in the expression of genes encoding antioxidant enzymes. Increased concentration of salt increased the activity of antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) in two popcorn genotypes but maximum activity was observed in the IAC125 genotype. Such enzymatic activities occur in order to maintain the levels of lipid peroxidation under salt stress, indicating that this genotype is tolerant to salinity conditions. The ZmAPX, ZmCAT, ZmSOD (Cu/Zn) and ZmSOD (Mn) genes increased their expression as salinity increased. The ZmSOD (Fe) gene was highly regulated in the IAC125 genotype under salt stress, but low regulation was observed in the UFM2 genotype, regardless of the salt concentration. The enhancement in tolerance against salt stress indicates that the genes involved in the antioxidative process are triggered by oxidative stress induced by abiotic stresses. These results showed that the popcorn varieties have different levels of salt tolerance due to the differential expression pattern of the antioxidant genes. The up-regulation of antioxidant enzymatic activity could lead to increased scavenging of excessive free radicals and reduce oxidative stress.

Conhecer a variabilidade genética existente entre cultivares de trigo (Triticum aestivum) é de grande importância para os programas de melhoramento. Desta forma, esse trabalho teve como objetivo utilizar marcadores ISSR para acessar a diversidade genética entre acessos contrastantes de trigo. Para isso, foram avaliados 32 acessos de trigo e um acesso de triticale provenientes do provenientes do Banco Ativo de Germoplasma de Trigo do Instituto de Desenvolvimento Rural do Paraná (IDR-Paraná). more » ... rtir de 10 primers de ISSR foram amplificados um total de 66 loci polimórficos com variação média de loci por primer de 6,6. A similaridade genética variou de 0,29 a 0,93, com média de 0,57. O dendrograma resultante mostrou 4 grupos, no qual observou-se que os genótipos LD 111202 e LD 111203 apresentaram alto índice de similaridade genética entre si. A análise bayesiana de agrupamento formou quatro grupos, parcialmente concordantes com o agrupamento hierárquico de UPGMA. Os marcadores ISSR foram capazes de discriminar acessos morfologicamente distintos e similares, além de fornecer informações consistentes sobre a variabilidade genética existente entre os genótipos de trigo que auxiliará nos programas de melhoramento genético.

Fungos micorrízicos arbusculares na tolerância a seca: resposta no metabolismo antioxidante de Ocimum basilicum L

O manjericão apresenta grande importância devido ao alto teor de substâncias bioativas presentes nas folhas, amplamente utilizada na medicina tradicional e na indústria farmacêutica. No entanto, como na maioria das plantas, mudanças fisiológicas e bioquímicas ocorrem em plantas de manjericão submetidas à seca. A fim de minimizar essas mudanças fungos micorrízicos arbusculares (FMAs) tem sido utilizado. Este estudo teve como objetivo avaliar o crescimento e a resposta antioxidante de plantas de
more » ... anjericão sob déficit hídrico e inoculados com Clarideoglomus etunicatum. O estudo foi feito em câmera de crescimento em plantas de manjericão inoculadas com FMAs e submetidas à seca após 30 dias após o transplante. Mudas inoculadas com FMA mostraram uma taxa de colonização de 28,66 e 32,79%. Plantas sem a inoculação e submetidas à seca apresentaram redução significativa no diâmetro do caule, comprimento da parte aérea e raiz e no acúmulo de biomassa da parte aérea e sistema radicular. Com a inoculação, houve uma melhora na resposta fisiológica das plantas sob estresse hídrico, referente a concentração de clorofila e o teor relativo de água. Grandes quantidades de prolina se acumularam nas folhas inoculadas e sob estresse, o que mostra o papel benéfico desse osmolitos em manjericão submetido à seca. As enzimas antioxidantes CAT e APX apresentaram maior atividade nas plantas expostas à seca e inoculadas com FMAs, o que reduziu o dano da membrana plasmática. A inoculação com FMAs em manjericão regulou positivamente a síntese de osmolitos e o metabolismo antioxidante promovendo tolerância à seca.

Genotoxicity of 22% Carbamide Peroxide Bleaching Agent on Oral Cells using the Micronucleus Technique

Several bleaching products and methods were introduced over the last years. However, the consequences of 22% carbamide peroxide on the genetic material of oral mucosal cells is uncertain. Aim: This study investigated the genotoxic effect of 22% carbamide peroxide on oral mucosal cells using the Micronucleus (MN) technique. Materials and Methods: Sixteen volunteers participated in the study. At-home bleaching was performed for 2 hours per day for 21 days. Cells were collected by scraping the
more » ... mucosa at four different evaluation times: Baseline (before treatment), 14 days, 21 days, and 52 days after the beginning of bleaching procedure. One thousand cells from each volunteer were examined and the frequencies of MN, binucleated cells, and metanuclear alterations were recorded. After preparing the slides, the microscopic fields were analysed from right to left, until a total of 1,000 cells were analysed. Data were analysed statistically using Kruskal Wallis test at 5% significance level. Results: There was a significant increase in the number of metanuclear alterations and binucleated cells between the baseline and day 21 (p<0.0001). The frequency of MN between the evaluation times was not significantly different (p=0.08). One month after bleaching, the alterations were similar to the baseline values. Conclusion: Although at-home bleaching using 22% carbamide peroxide can cause temporary effects on mucosal cells, the alterations disappear one month after finishing the bleaching treatment.

Mitochondria are the energy-generating hubs of the cell. In spite of considerable advances, our understanding of the factors that regulate the molecular circuits that govern mitochondrial function remains incomplete. Using a genome-wide functional screen, we identify the poorly characterized protein Zinc finger CCCH-type containing 10 (Zc3h10) as regulator of mitochondrial physiology. more » ... ion of myoblasts into myotubes. Zc3h10 overexpression boosts mitochondrial function and promotes myoblast differentiation, while the depletion of Zc3h10 results in impaired myoblast differentiation, mitochondrial dysfunction, reduced expression of electron transport chain (ETC) subunits, and blunted TCA cycle flux. Notably, we have identified a loss-of-function mutation of Zc3h10 in humans (Tyr105 to Cys105) that is associated with increased body mass index, fat mass, fasting glucose, and triglycerides. Isolated peripheral blood mononuclear cells from individuals homozygotic for Cys105 display reduced oxygen consumption rate, diminished expression of some ETC subunits, and decreased levels of some TCA cycle metabolites, which all together derive in mitochondrial dysfunction. Taken together, our study identifies Zc3h10 as a novel mitochondrial regulator.

O déficit hídrico altera o crescimento, metabolismo oxidativo e biossíntese de fenilpropanóides em Ocimum basilicum L

Mudanças fisiológicas e bioquímicas ocorrem quando as plantas são submetidas ao estresse hídrico. Portanto, o objetivo desse trabalho foi avaliar o crescimento, a resposta antioxidante, o rendimento e a composição do óleo essencial (OE) do manjericão (Ocimum basilicum L.) submetidas ao déficit hídrico. O experimento foi conduzido em casa de vegetação sob três regimes hídricos: 100-90% (controle), 70-60% (estresse moderado), 40-30% (estresse severo) da capacidade de campo por 50 dias. Plantas
more » ... metidas ao estresse hídrico severo apresentaram reduções significativas na massa fresca e altura. O teor relativo de água nas folhas diminuiu com o aumento da severidade do estresse. Observamos um aumento na atividade de enzimas: superóxido dismutase (SOD), ascorbato peroxidase (APX) e catalase (CAT) em plantas submetidas ao déficit hídrico. As plantas submetidas ao estresse moderado e serevo apresentaram maior rendimento do OE, além de alterar sua composição química. Um total de 54 compostos foram identificados no OE das folhas de manjericão. Os fenilpropanóides aumentaram com o déficit hídrico e podem constituir um sistema antioxidante secundário, que pode auxiliar o sistema de defesa antioxidante primário, a fim de manter os níveis de peróxido de hidrogênio nas células em concentrações sub-letais para as plantas de manjericão.
