Effect of puerarin on human choriocarcinoma cells

AbstractObjective: To discuss the effect of puerarin on human choriocarcinoma cells. Methods: Survival rates under puerarin monotherapy, fluorouracil (5-FU) monotherapy and puerarin in combination with 5-FU were detected by MTT assay. Apoptotic morphology was observed with Hoechst 33258 staining. Apoptosis rates were detected with flow cytometry. Expressions of AKT, mechanistic target of rapamycin (mTOR), and P70S6K mRNAs and phosphorylated proteins were detected by RT-PCR and Western blot.
more &raquo; ... r-bearing mice were administered puerarin and puerarin+5-FU, and serum levels of β-human chorionic gonadotropin (β-HCG) were measured. Results: Proliferation inhibition and apoptosis rates of JEG-3 cells were positively correlated with puerarin concentration, which increased in the puerarin+5-FU group. Expression levels of AKT, mTOR, P70S6K mRNAs, and phosphorylated proteins decreased significantly after action of puerarin at different concentrations. With increasing puerarin concentration, expression of cleaved-caspase-3 in JEG-3 cells increased, whereas that of Bcl-2 decreased. Puerarin significantly inhibited tumor growth in choriocarcinoma-bearing SCID mice. Serum β-HCG levels were significantly lower than those of control group after administration. Magnitude of β-HCG decline was positively correlated with concentration. Conclusion: Puerarin+5-FU inhibited proliferation of JEG-3 choriocarcinoma cells and promoted their apoptosis, being associated with the mTOR signaling pathway.

A Livelihood Resilience Measurement Framework for Dam-Induced Displacement and Resettlement

The concepts of resilience and livelihood resilience are growing in prominence with water resource development that aims to measure and build resilience to specific disturbances and shocks. However, practical frameworks to measure livelihood resilience are needed, not just a theoretical framework. In this paper, we introduce the livelihood resilience measurement (LRM) framework, which draws on Hooke's law; use the state vector method to calculate livelihood resilience scores; and test the
more &raquo; ... iveness of the method by correlation analysis. We illustrate the framework by using it to measure livelihood resilience in Henan Province, China, and assess the strategies that build livelihood resilience. The advantages and limitations of the framework are explored and discussed by drawing on empirical examples. Besides illustrating how to apply the LRM framework in a practical case, we discuss how to communicate with stakeholders to identify and strengthen the factors that build resilience. In this study, land ownership, housing and property value, and emergency cost are the most significant of these factors. Thus, the LRM framework has the potential to help reservoir-affected families protect their livelihood capital and to help governments improve social welfare. It can thus serve as a tool for monitoring and improving the effectiveness of policies and practices aimed at building livelihood resilience.

Conjugation of Inulin Improves Anti-Biofilm Activity of Chitosan

Bacteria biofilm helps bacteria prevent phagocytosis during infection and increase resistance to antibiotics. Staphylococcus aureus is a Gram-positive pathogenic bacterium and is tightly associated with biofilm-related infections, which have led to great threat to human health. Chitosan, the only cationic polysaccharide in nature, has been demonstrated to have antimicrobial and anti-biofilm activities, which, however, require a relative high dosage of chitosan. more &raquo; ... rther restricts its applications on anti-infection therapy. Inulins are a group of polysaccharides produced by many types of plants, and are widely used in processed foods. Compared to chitosan, inulin is very soluble in water and possesses a mild antibacterial activity against certain pathogenic bacteria. In order to develop an effective strategy to treat biofilm-related infections, we introduce a method by covalent conjugation of inulin to chitosan. The physicochemical characterization of the inulin-chitosan conjugate was assayed, and the anti-biofilm activity was evaluated against S. aureus biofilm. The results indicated that, as compared to chitosan, this novel polysaccharide-polysaccharide conjugate significantly enhanced activities against S. aureus either in a biofilm or planktonic state. Of note, the conjugate also showed a broad spectrum anti-biofilm activity on different bacteria strains and low cellular toxicity to mammalian cells. These results suggested that chitosan conjugation of inulin was a viable strategy for treatment against biofilm-related infections. This finding may further spread the application of natural polysaccharides on treatments of infectious disease.

Infrastructure Investment and Regional Economic Growth: Evidence from Yangtze River Economic Zone

To better understand the impacts of infrastructure investment on economic growth and, we used Cobb-Douglas production function model to develop the stock of public infrastructure capital into the economic growth model. It applies spatial panel data model effect analysis to statistical data of the Yangtze River Economic Zone with 131 cities from 2003 to 2016 and investigates the relationship between different types of public infrastructure capital stock and regional economic growth in different

The empirical results show that (1) the economic growth of the cities in the Yangtze River Economic Zone has characteristics of significant spatial dependence, the degree and significance of spatial dependence are gradually declining, and the spatial agglomeration of the economic growth in the cities in the region is relatively stable. (2) Different types of public infrastructure capital stock have distinct spatial effects on regional economic growth. The capital stock of energy infrastructure significantly promotes global economic growth. The capital stock of transportation infrastructure significantly stimulates the local economic growth and inhibits the economic growth of the adjacent areas. The capital stock of water-related infrastructure restrains local economic growth and promotes economic growth in adjacent areas. These findings indicate that increasing investment in public infrastructure development in the Yangtze River Economic Zone remains an effective measure to promote regional economic growth. Under the premise of limited resources, taking full account of the effects of various types of investment can promote the mutual benefit of the economies in the region and effectively achieve the strategic objectives for the Yangtze River Economic Zone.

In the analysis of equipment failure, the data before and after fault and before and after fault are analyzed. The abnormal state of equipment is associated with the data before and after the fault. Therefore, by analyzing the characteristics of the fault data and the equipment before and after the fault, a bidirectional recurrent neural network model based on LSTM is constructed. The method designed in this paper can not only improve the efficiency and speed of collection, but also can compare and collect fault information. The overall operation state of the power unit is improved accurately. The research results show that the company's low-voltage user acquisition success rate has reached more than 99%. With the increase of time, the fault information collection efficiency can approach 99%. It shows that the function of this research system is better, the economic loss of the company is reduced, and the management is optimized.

A voltage-mode ultra low power four quadrant analog multiplier using subthreshold PMOS is presented in this paper. PMOS substract cell and combiner cell operating in the subthreshold region are used to lower the voltage-mode multiplier power consumption. Simulation results of the multiplier demonstrate a linearity error of 0.8%, a maximum THD of 4%, a −3 dB bandwidth of 1.4 MHz, and a power consumption of 77 nW with 100 fF load capacitor at a supply voltage of 0.6 V using a TSMC 0.18 um CMOS process.

Lin28B is a RNA-binding protein that inhibits the let-7 microRNA family and acts as an oncogene in various human malignant diseases. more &raquo; ... racteristics. Breast cancer patients previously diagnosed with invasive ductal carcinomas were enrolled in this study. All cases went through surgical procedures as the initial treatment. The characteristics of every case were collected, including tumor size, pathologic grade, metastatic lymphoid nodes, and estrogen receptor a (ERa), progesterone receptor (PR), and HER2 status. The immunostaining was scored by two independent investigators. Eighty-three (43.7%) of 190 cases showed positive expression of Lin28B. Lin28B immunostaining was increased in tumors compared with the adjacent tissues. Overexpression of Lin28B was linked to poor differentiation, advanced-stage disease, and Ki67-positive status (all p < 0.05). Besides, Lin28B expression was significantly different among breast cancer subtypes. This study addresses the role of Lin28B in breast cancers and provides insight of its predictive effects in disease development.

Genomic and Transcriptomic Dissection of the Large-Effect Loci Controlling Drought-Responsive Agronomic Traits in Wheat

Drought tolerance is one of the most important targets for wheat breeding. Previous population genetics studies have uncovered 20 large-effect quantitative trait loci (QTLs) that contribute to stress-responsive agronomic traits. Here, we identified 19,035,814 single nucleotide polymorphisms and 719,049 insertion/deletion variations in the genomes of two popular winter wheat cultivars, Lu-Mai 14 and Han-Xuan 10, using a whole-genome re-sequencing assay. more &raquo; ... carried by protein-coding genes, such as CCA1/LHY, AGO1, ABI3/VP1, EIN3, TPP, and ARFs. We carried out a time-course abscisic acid (ABA)-treatment experiment and profiled 61,251 expressed genes in the roots using a strand-specific RNA sequencing approach. A large number of genes showed time-point specific and/or cultivar-preferential responsive expression patterns. Gene ontology enrichment analysis revealed that ABA-responsive genes were associated with stress-related functions. Among the 20 QTLs, we uncovered 306 expressed genes with high- and/or moderate-effect variations and 472 differentially expressed genes. Detailed analysis and verification of the homozygous genomic variations in the candidate genes encoding sulfotransferase, proteinase, kinase, nitrate transporter, and transcription factors suggested previously unexpected pathways associated with abiotic stress responses in wheat.

It builds the system in the MATLAB/Simulink simulation environment mathematical model. We use the critical proportionality method to determine many critical parameters in the control system, tune the proportional coefficient, integral time, and other physical quantities in the PID controller, and analyze the proportional control, proportional-integral control, PID controls the respective dynamic response characteristics of these three different control systems. The simulation and comparative analysis results show that: compared with the other two control methods, PID control can effectively weaken the heat storage system oscillation caused by external disturbance, its dynamic response speed is faster, the adjustment time is shorter, and it can meet the requirements of operational stability. The paper adopts PID control, which reduces the control difficulty of the trough solar heat storage system and improves the adaptability to changes in external meteorological resources. The research results have particular guiding significance at the academic and engineering levels.
