Synergic Activity Against MCF-7 Breast Cancer Cell Growth of Nanocurcumin-Encapsulated and Cisplatin-Complexed Nanogels

Nanogel-based systems loaded with single anticancer drugs display miscellaneous effectiveness in tumor remission, gradually circumventing mutation and resistance in chemotherapy. Hence, the existence of dual-drug delivered nano-sized systems has been contemporaneous with drug development and preceded the conventional-dose chemotherapy. Among outstanding synergistic drug nanoplatforms, thermosensitive copolymer heparin-Pluronic F127 (Hep-F127) co-delivering cisplatin (CDDP) and curcumins (Cur)
more » ... ep-F127/CDDP/Cur) has emerged as a notable candidate for temperature-responsive drug delivery. The procedure was based on the entrapment of curcumin into the hydrophobic core of bio-degradable co-polymer Hep-F127 while the hydrophilic drug CDDP subsequently conjugated to the backbone heparin to form the core-shell structure. The copolymer was characterized by Fourier transform infrared (FT-IR) spectrophotometry, Transmission Electron Microscopy (TEM), and Dynamic Light Scattering (DLS), to corroborate the successful synthesis and via HPLC along with AES-ICP to evaluate the high drug loading along with a controllable release from the nano-gels. A well-defined nano-shell with size in the 129.3 ± 3.8 nm size range could enhance higher the efficacy of the conjugated-CDDP to Hep-F127 than that of single doses. Moreover, the considerable amount of dual-drug released from thermosensitive nanogels between different conditions (pH = 7.4 and pH = 5.5) in comparison to CDDP from Hep-F127 partially indicated the significantly anti-proliferative ability of Hep-F127/CDDP/Cur to the MCF-7 cell line. Remarkably, drug testing in a xenograft model elucidates the intricate synergism of co-delivery in suppressing tumor growth, which remedies some of the problems affecting in cancer chemotherapy.

In this study, the optimal conditions for the spray drying from Trametes versicolor (V.C.) extracts were determined using response surface method/technique (RSM). Forty-three myxobacterial strains were isolated and identified from 20 soil samples collected in Vietnam. The information on morphological characteristics and 16S ribosomal gene sequencing showed that these strains were designated to seven genera belonging to Angiococcus, Archangium, Chondromyces, Corallococcus, Cystobacter, Melittangium, and Myxococcus, in suborder Cystobacterineae and Sorangiineae. more » ... isolates. Myxobacteria were cultured, and crude extracts were obtained after 10-day fermentation in P-medium in the presence of the Amberlite XAD 16N adsorbent resin. Elution was carried out with acetone and methanol to obtain the crude extracts. Evaluation of antioxidant activity used the DPPH and ABTS assay, the minimum inhibitory concentration values were determined by the microdilution method. The total extract from CT21 had the highest total antioxidant activity (IC50 = 52.34 +/- 1.47 mcg/mL, 30.28 +/- 0.74 mcg/mL for DPPH and ABTS assays, respectively). The other potential strain was TG131 and GL41 that IC50 values were 40.28 +/- 1.13 and 57.24 +/- 1.52 mcg/mL, respectively (by the DPPH method), and 48.35 +/- 0.58 and 42.76 +/- 0.50 mcg/mL, respectively (by the ABTS method). Interestingly, 100% isolated myxobacterial strains show inhibitory activity against at least one of the tested microorganisms. The potential antimicrobial strain was GL41, which inhibited all tested microorganisms, and the MIC values were 1 mcg/mL against MRSA, MSSA, S. faecalis, C. albicans, and A. niger. The highest active strains were members of Myxococcus sp. genus.

The under-representation of Vietnamese ethnic groups in existing genetic databases and studies have undermined our understanding of the genetic variations and associated traits or diseases in the population. Cost and technology limitations remain the challenges in performing large-scale genome sequencing projects in Vietnam and many developing countries. Non-invasive prenatal testing (NIPT) data offers an alternative untapped resource to study genetic variations in the Vietnamese population.
more » ... ults: We analyzed the low-coverage genomes of 2,683 pregnant Vietnamese women using their NIPT data and identified a comprehensive set of 8,054,515 single-nucleotide polymorphisms, among which 8.2% were new to the Vietnamese population. Our study also revealed 24,487 disease-associated genetic variants and their allele frequency distribution, especially five pathogenic variants for prevalent genetic disorders in Vietnam. We also observed major discrepancies in the allele frequency distribution of disease-associated genetic variants between the Vietnamese and other populations, thus highlighting a need for genome-wide association studies dedicated to the Vietnamese population. Conclusions: We have demonstrated a successful analysis of NIPT data to reconstruct the Vietnamese genetic profiles. This application provides a powerful yet cost-effective approach for large-scale population genetic studies.

Encapsulating gold nanoparticles in zeolitic imidazolate framework crystal for novel optical response

We investigated optical properties of Au nanoparticles (Au NPs) loaded into the crystal of a porous material ZIF-8, abbreviated as Au@ZIF-8, by a diffuse reflectance spectroscopy. Au@ZIF-8 was synthesized by a reaction of ZIF-8 precursor with PVP-encapsulated-Au-2 NPs in methanol solution. The concentration of Au NPs in an aqueous solution of the ZIF-8 precursor was controlled from 0.05 mL to 1.0 mL. more » ... der X-ray diffraction (PXRD) analysis, Raman spectroscopy, SEM, and TEM images. The upper limit of concentration for Au NPs positioned inside ZIF-8 crystal is approximately 0.05 mL. At higher concentrations, the crystal size of ZIF-8 becomes smaller; and some Au NPs are observed to be located outside the framework. The optical spectra of Au@ZIF-8 exhibit two main absorption bands at ≈520 nm and ≈820 nm. The former of which is attributed to the surface plasmon resonance of Au NPs, while the band at ≈820 nm can be assigned to the longitudinal mode and/or collective plasmonic modes, as long as the particles are closely aggregated. Finite-Difference Time-Domain (FDTD) calculation for extinction cross-section spectrum for a BCC arrangement of Au NPs with the radius of 5 nm and the center-to-center distance of 13 nm was utilized to confirm the observation of new optical response of Au@ZIF-8. The calculation is in rather good agreement with the experimental data showing two main bands at ≈500 nm and ≈800 nm. This work suggests a way to create a magnetic material in optical frequency in case of locating Au NPs periodically in the pores of ZIF-8 or more generally in the metal-organic frameworks (MOFs).

Effect of the Optimize Heart Failure Care Program on clinical and patient outcomes – The pilot implementation in Vietnam

The Ho-Chi-Minh-city Heart Institute in Vietnam took part in the Optimize Heart Failure (OHF) Care Program, designed to improve outcomes following heart failure (HF) hospitalization by increasing patient awareness and optimizing HF treatment. HF patients hospitalized with left ventricular ejection-fraction (LVEF) <50% were included. Patients received guideline-recommended HF treatment and education. Clinical signs, treatments and outcomes were assessed at admission, discharge, 2 and 6 months
more » ... , M6). Patients' knowledge and practice were assessed at M6 by telephone survey. 257 patients were included. Between admission and M2 and M6, heart rate decreased significantly, and clinical symptoms improved significantly. LVEF increased significantly from admission to M6. 85% to 99% of patients received education. At M6, 45% to 78% of patients acquired knowledge and adhered to practice regarding diet, exercise, weight control, and detection of worsening symptoms. High use of renin-angiotensin-aldosterone-system inhibitors (91%), mineralocorticoid-receptor-antagonists (77%) and diuretics (85%) was noted at discharge. Beta-blocker and ivabradine use was less frequent at discharge but increased significantly at M6 (from 33% to 51% and from 9% to 20%, respectively, p < 0.001). There were no in-hospital deaths. Readmission rates at 30 and 60 days after discharge were 8.3% and 12.5%, respectively. Mortality rates at 30 days, 60 days and 6 months were 1.2%, 2.5% and 6.4%, respectively. The OHF Care Program could be implemented in Vietnam without difficulty and was associated with high usage of guideline-recommended drug therapy. Although education was delivered, patient knowledge and practice could be further improved at M6 after discharge.

more » ... ion-desorption isotherms, dynamic light scattering (DLS), thermogravimetric analysis (TGA), and Fourier transform infrared spectroscopy (FTIR) were employed. The size of HMSN nanoparticles in aqueous solution averaged 134.0 ± 0.3 nm, which could be adjusted by minor changes during synthesis, whereas that of PNS nanoparticles was 63.4 ± 0.6 nm. In addition, the encapsulation of RhB into HMSN nanoparticles to form RhB-loaded nanocarriers (RhB/HMSN) was successful, achieving high loading efficiency (51.67% ± 0.11%). This was significantly higher than that of RhB-loaded PNS (RhB/PNS) (12.24% ± 0.24%). Similarly, RhB/HMSN also possessed a higher RhB loading content (10.44% ± 0.02%) compared to RhB/PNS (2.90% ± 0.05%). From those results, it is suggested that prepared HMSN nanocarriers may act as high-capacity carriers in drug delivery applications.
