Stimulation of phagocytosis in mouse peritoneal macrophages by orexin-B and orexin-A

To defi ne the effects of feeding and sleep regulating peptides, orexins, in immunocompetent cells, the effects of orexin-A and orexin-B on phagocytosis in mouse peritoneal macrophages were examined. Orexin-B induced an enhancement of phagocytosis in a dose-dependent manner. Orexin-A is less effective than orexin-B. Even in Ca 2+ -free solutions, phagocytosis was enhanced by orexin-B. The potassium channel blocker quinine inhibited the enhanced phagocytosis by orexin-B; 4-aminopyridine and
more » ... ethylammonium suppressed phagocytosis less effectively. These results suggest that orexins can enhance the phagocytosis of macrophages mediated by potassium channels.

The aim of this study was to grasp the injury situation of Japanese female rugby players in 2020-2021 by network analysis with psychological condition under COVID19 (June 1-30, 2021). The number of respondents was 302. 60% reported serious injuries. The parts were ankle, knee, shoulder, head, etc. Symptoms are ligament / bone / muscle damage or concussion. The caused plays were tackle/tackled, running with no physical contact, etc. There were 39 head injuries. Some ones played while continuing
more » ... ong-term treatment such as knees and ankles. Approximate 40 % of players injured once were injured twice again, and approximate 40% of players injured twice were injured a third time. 30% of all respondents had some chronic symptoms. Network analysis revealed a structure with multiple chronic symptoms (head, shoulders, knees, ankles), a structure of multiple parts of injuries in a single play, and a structure in which the same parts were repeatedly injured in a year. Some players suffered concussions after knee injuries. It suggested the possibility that the physical safety management skills to protect the head might be inferior due to insufficient recovery of knee function as a negative injury chain structure.

After the first World Cup 1987, rugby undergoes rule revisions aimed at more dynamic games. There have been some analyses of the KPIs (Key Performance Indicators) so far, but not many ones as detailed dynamic network structure of tactics concerned with selected attack and defense areas, plays, and human resources. In current study, the tactics for try in Rugby World Cup 2019 was investigated by network centrality, core-periphery analysis and correspondence analysis. Bootstrap test and ROC
more » ... is were used to validate the data of try contribution structure. The average score of try balance of final 8 teams was "3.94" and that of "not win" teams was "-2.23". We categorized these indices into team performance, and tested Monte Carlo methods with bootstrap hypothesis testing to assess the standardized values. Furthermore, to test the precision of sensitivity and specificity of standardized try balance values, the Area Under the Curve (AUC) of the receiver-operator curve (ROC) analysis was executed. In final 8 stage, the feature of tactics for try in first 20 minutes and last 20 minutes were analyzed. The results suggested the tactics of "attack channel diversity" in first 20 minutes and tactics of "defense and substitute diversity" in last 20 minutes. In addition, network correspondence analysis of the top 4 teams' performance in the tournament yielded interesting results regarding tactics of the attack and defense methods, and of the transition of human resources.

Cytomegalovirus (CMV) infection exacerbates ulcerative colitis (UC) refractory to immunosuppressive therapies. However, the underlying UC remained active in some UC patients, despite the fact that CMV-DNA in colonic mucosa became negative after antiviral therapy. Therefore, new therapeutic strategies for UC patients concomitant with CMV infection in mucosa are required. Aims: The aim of this study was to evaluate the effect and safety of granulocyte-monocyte adsorption apheresis (GMA) in UC
more » ... ents positive for CMV infection after antiviral therapy. Methods: From October 2003 to December 2008, 64 patients with UC refractory to immunosuppressive therapies (IMT), including steroids and immunomodulators, were enrolled in this retrospective, observational, multicenter study, which was reviewed and approved by the Institutional Review Board of Kyoto University. CMV infection was investigated by three methods (histologic examination, CMV antigenemia, and polymerase chain reaction). We investigated clinical outcomes of GMA and IMT after 2weeks of treatment with ganciclovir. Results: Thirty-one (48.4%) of 64 patients with UC refractory to IMT were positive for CMV. Of 31 patients, 4 (12.9%) underwent colectomy. Twenty-seven patients (87.1%) underwent antiviral therapy. Seven of 27 patients achieved remission following antiviral therapy alone. Of the remaining 20 patients that did not achieve remission despite the disappearance of CMV-DNA, 11 and 9 patients were treated with additional GMA (GMA group) and IMT (IMT group), respectively. Nine of 11 patients (GMA group) achieved remission and 2 underwent colectomy. Four of 9 patients (IMT group) achieved remission and 5 underwent colectomy. CMV-DNA was not detected in 11 Yoshino et al. 4 patients after GMA, but it was detected again in all 5 patients of the IMT group, that underwent colectomy. Total colectomy rate in UC patients positive for CMV was 35.5% (11/31). In addition, colectomy-free survival in the CMV relapse (+) group was estimated to be 12.9% at 65 months, while that in the CMV relapse (-) group was estimated to be 100% at 60 months. Conclusion: The colectomy ratio tends to be high in refractory UC patients with recurrent CMV reactivation or infection. Therefore, GMA might be a safe and effective treatment for UC patients positive for CMV because it does not induce avoiding CMV reactivation.

The authors have developed the vegetation model which considers soil nutrient cycle. In this study, at first, we improve the vegetation model by updating of the flush mechanism of trees and addition of the mechanism that the trees require TN from atmosphere. Then, we simulated the change of the vegetation by the soil nutrient cycle on a sandbar. As a result, we confirmed that the robinia decrease when the soil nutrient increase.
