Amplification of L-myc Oncogene in Malignant Meningioma

Amplification of L-myc oncogene was noticed in a malignant meningioma originating from the right sphenoidal wing of a 54-year-old female. The patient underwent three surgical resections plus radiotherapy over a period of 11 years and then the growth rate of the tumor became much greater with a severely invasive appearance. Using Southern blot hybridization, L-myc amplification was examined on the specimen resected at the fourth operation. As a result , approximately five-fold amplification was
more &raquo; ... onfirmed, which has not been previously reported except for that in a small cell carcinoma of the lung. This result may suggest that L-myc amplification is responsible to some extent for the malignant transformation in this meningioma.

Sufficient therapeutic effect of cryopreserved frozen adipose-derived regenerative cells on burn wounds [article]

The purpose of this study was to evaluate whether cryopreserved (frozen) adipose-derived regenerative cells (ADRCs) have a therapeutic effect on burn wound healing as well as freshly isolated (fresh) ADRCs. Full thickness burns were created on dorsum of nude mice and burn wound was excised. The wound was covered by artificial dermis with; (i) fresh ADRCs, (ii) frozen ADRCs, and (iii) PBS (control). The assessment for wound healing was performed by morphological, histopathological and
more &raquo; ... chemical analyses. In vivo analyses exhibited the significant therapeutic effect of frozen ADRCs on burn wound healing up to the similar or higher level of fresh ADRCs. There were significant differences of wound closure, epithelized tissue thickness, and neovascularization between the treatment groups and control group. Although there was no significant difference of therapeutic efficacy between fresh ADRC group and frozen ADRC group, frozen ADRCs improved burn wound healing process in dermal regeneration with increased great type I collagen synthesis compared with fresh ADRCs. These findings indicate that frozen ADRCs allow us to apply not only quickly but also for multiple times, and the cryopreserved ADRCs could therefore be useful for the treatment of burn wounds in clinical settings.

We conducted enclosure experiments in a shallow eutrophic lake, in which a biomass gradient of the filter-feeding planktivore, silver carp, Hypophthalmichthys molitrix Valenciennes, was created, and subsequent community changes in both zooplankton and phytoplankton were examined. 2. During a summer experiment, a bloom of Anabaena flos-aquae developed (» 8000 cells mL ±1 ) solely in an enclosure without silver carp. Concurrent with, or slightly preceding the Anabaena bloom, the number of
more &raquo; ... er species and their abundance increased from seven to twelve species (1700±14 400 organisms L ±1 ) after the bloom in this fish-free enclosure. Protozoans and bacteria were generally insensitive to the gradient of silver carp biomass. 3. During an autumn experiment, on the other hand, large herbivorous crustaceans were more efficient than silver carp in suppressing the algae, partly because the lower water temperature (» 24°C) inhibited active feeding of this warm-water fish and also formation of algal colonies. Heterotrophic nanoflagellate and bacterial densities were also influenced negatively by the crustaceans. 4. Correspondence analysis (CA) was applied to the weekly community data of zooplankton and phytoplankton. A major effect detected in the zooplankton community was the presence/absence of silver carp rather than the biomass of silver carp, whereas that in the phytoplankton community was the fish biomass before the Anabaena bloom, but shifted to the presence/absence of the fish after the bloom.

β1-Adrenergic Receptor Recycles Via a Membranous Organelle, Recycling Endosome, by Binding with Sorting Nexin27

In cardiomyocytes, b 1 -adrenergic receptor (b 1 -AR) plays an important role in regulating cardiac functions. Upon continuous ligand stimulation, b 1 -AR is internalized and mostly recycled back to the plasma membrane (PM). The recycling endosome (RE) is one of the membranous organelles involved in the protein recycling pathway. To determine whether RE is involved in the internalization of b 1 -AR upon ligand stimulation, we evaluated the localization of b 1 -AR after stimulation with a
more &raquo; ... st, isoproterenol (Iso), in b 1 -AR-transfected COS-1 cells. After 30 min of Iso treatment and cell surface labeling with the appropriate antibodies, b 1 -AR was internalized from PM and translocated into the perinuclear region, the same location as the transferrin receptor, an RE marker. We then evaluated whether sorting nexin 27 (SNX27) participated in the b 1 -AR recycling pathway. When b 1 -AR and SNX27 were coexpressed, b 1 -AR coimmunoprecipitated with SNX27. In addition, shRNA-mediated silencing of SNX27 compromised b 1 -AR recycling and enhanced its delivery into lysosome. Overall, b 1 -AR on PM was internalized into RE upon Iso stimulation and recycled by RE through binding with SNX27 in COS-1 cells.

Abstracts--- Ba ferrite/Mn-Zn ferrite multilayered thin films have been investigated to decrease high coercive force of Ba ferrite single layer films, which is suitable for perpendicular magnetic recording media. Specimen films were deposited by using Facing Targets Sputtering apparatus. Thickness of Ba ferrite layer and Mn-Zn ferrite, which ratio was fixed at 5:2, and partial oxygen pressure were changed, and crystal structure and magnetic properties of such films have been investigated. As a
more &raquo; ... esult, all specimen films have perpendicular magnetic anisotropy. It was found that perpendicular coercive force could be controlled from 2kOe to 6000e by adjusting each layer thickness. Ba ferrite 25A/ Mn-Zn ferrite lOA multilayered film prepared without oxygen gas showed saturation magnetization Ms of about 450emu/cc and perpendicular coercive force Hc.!. of about 6000e, which is suitable value for perpendicular magnetic recording with ultra high density.

Sufficient therapeutic effect of cryopreserved frozen adipose-derived regenerative cells on burn wounds

The purpose of this study was to evaluate whether cryopreserved (frozen) adipose-derived regenerative cells (ADRCs) have a therapeutic effect on burn wound healing as well as freshly isolated (fresh) ADRCs. Full thickness burns were created on dorsum of nude mice and burn wound was excised. The wound was covered by artificial dermis with; (i) fresh ADRCs, (ii) frozen ADRCs, and (iii) PBS (control). The assessment for wound healing was performed by morphological, histopathological and
more &raquo; ... chemical analyses. In vivo analyses exhibited the significant therapeutic effect of frozen ADRCs on burn wound healing up to the similar or higher level of fresh ADRCs. There were significant differences of wound closure, epithelized tissue thickness, and neovascularization between the treatment groups and control group. Although there was no significant difference of therapeutic efficacy between fresh ADRC group and frozen ADRC group, frozen ADRCs improved burn wound healing process in dermal regeneration with increased great type I collagen synthesis compared with fresh ADRCs. These findings indicate that frozen ADRCs allow us to apply not only quickly but also for multiple times, and the cryopreserved ADRCs could therefore be useful for the treatment of burn wounds in clinical settings.

We investigated the anti-tumoral effect of intratumoral injection using nude mice with subcutaneously transplantated MKN-45 gastric cancer followed by exposure to red light in order to avoid phototoxic reaction after intravenous administration of hematoporphyrin derivatives (HpD). Histopathological findings revealed that necrosis was uniform in the group administered intraperitoneally but it was not so in the group administered intratumorally.
