A note on the top-down and bottom-up gradient functions over a forested site

The dimensionless bottom-up and top-down gradient functions in the convective boundary layer (CBL) are evaluated utilizing long-term well-calibrated carbon dioxide mixing ratio and flux measurements from multiple levels of a 447-m tall tower over a forested area in northern Wisconsin, USA. The estimated bottomup and top-down functions are qualitatively consistent with those from large-eddy simulation (LES) results and theoretical expectations. Newly fitted gradient functions are proposed based
more &raquo; ... n observations for this forested site. The integrated bottom-up function over the lowest 4% of the CBL depth estimated from the tower data is about five times larger than that from LES results for a 'with-canopy' case, and is smaller than that from LES results for a 'no-canopy' case by a factor of 0.7. We discuss the uncertainty in the evaluated gradient functions due to stability, wind direction, and uncertainty in the entrainment flux and show that while all of these have a significant impact on the gradient functions, none can explain the differences between the modelled and observed functions. The effects of canopy features and atmospheric stability may need to be considered in the gradient function relations.

Histone deacetylase inhibitors induce growth suppression and cell death in human rhabdomyosarcoma in vitro

A group of histone deacetylase inhibitors has been shown to be effective in suppressing the growth of a variety of transformed cell lines in vitro and in vivo. The effects of two of these agents, suberoylanilide hydroxamic acid (SAHA) and suberoyl-3-aminopyridineamide hydroxamic acid (pyroxamide), were investigated for their growth-suppressive effects on rhabdomyosarcoma (RMS) cells. Dose-response experiments of two RMS cell lines, RD (embryonal) and RH30B (alveolar), were performed with SAHA
more &raquo; ... .25-3.0 micro M) and pyroxamide (1.25-20.0 micro M). Both agents caused a dose-dependent decrease in viable cell number and an increase in percentage of dead cells over time. Exposure of the RMS cells to SAHA and pyroxamide resulted in an accumulation of acetylated histones with increasing doses by Western blot analysis. Additionally, there was an induction of p21/WAF1 at 15 and 24 h when the cells were cultured with SAHA (2.0 micro M) or pyroxamide (20.0 micro M), concentrations that were tested because they successfully induced inhibition of cell growth and initiated cell death in both RMS cell lines. An increase in nuclei with hypodiploid or sub-G(1) fraction was found by flow cytometry with increasing doses of both SAHA (0.25-3.0 micro M) and pyroxamide (1.25-20.0 micro M) over time. This finding is consistent with DNA fragmentation and cell death by apoptosis. SAHA and pyroxamide induce growth suppression and cell death in human RMS in vitro. Accumulation of acetylated histones and induction of p21/WAF1 expression are observed in cells exposed to either agent.

Surface layer CO2 budget and advective contributions to measurements of net ecosystem–atmosphere exchange of CO2

The aim of this case study is to examine the surface layer CO 2 budget and possible impacts of advection on the 20 km scale on estimates of net ecosystem-atmosphere exchange (NEE) of CO 2 under windy conditions. The advection terms in the CO 2 mixing ratio conservation equation are computed using data from two flux towers 20 km apart in a forested area in Wisconsin, USA. The data are analyzed when the winds blow continuously from the southeast (SE) or from the northwest (NW) under windy
more &raquo; ... ns for 5 h or longer during the months of May through September of 1999 and 2000. Compared to the vertical turbulent flux, the contribution of horizontal flux divergence is negligible on the scale of 20 km at all times. The contributions of both horizontal and vertical advection terms to NEE estimates are negligible in the day. The mean nocturnal vertical and horizontal components of the advective fluxes have opposite signs and are of the same order of magnitude. Considering only one of the two advection terms in NEE calculation would be inappropriate. On the spatial scale specified, the contribution of horizontal advection is negative and can be 10% of NEE at night under sustained SE winds. The contribution of vertical advection is positive and is about 20% of NEE for the same wind direction. The contribution of nocturnal advection is negligible for most cases with sustained NW winds. The evaluation of the advective effects is still incomplete only from this dataset and significant contribution of advection on the scales smaller than 20 km is not ruled out because the 20-km scale may not be the primary scale of the heterogeneous distributions of land cover and soils. Nevertheless, the analyses suggest that significant errors due to the neglect of the impacts of the land cover and soil heterogeneity at larger scales (than local) are possible in one-dimensional eddy-covariance NEE measurements under windy conditions; these measurements are usually selected to describe nighttime NEE at this site and, therefore, the likely errors may deserve attention. The analyses also suggest that impacts of both advection terms within the surface layer on NEE estimates from the atmospheric boundary budget method could be negligible in the day but significant at night. More comprehensive experiments are needed to completely assess the advection issue. #
