Structural Implications of Siglec-5-Mediated Sialoglycan Recognition
2008
Journal of Molecular Biology
Sialic acid Ig-like binding lectins are important mediators of recognition and signaling events among myeloid cells. To investigate the molecular mechanism underlying Siglec functions, we have determined the crystal structure of the two N-terminal extracellular domains of a human myeloid cell inhibitory receptor Siglec-5 (CD170) and its complexes with two sialylated carbohydrates. The native structure revealed an unusual conformation of the CC′ ligand specificity loop and a unique interdomain

... sulfide bond. The α(2,3)-sialyllactose and α(2,6)-sialyllactose complexed structures showed a conserved sialic acid recognition motif that involves both Arg 124 and a portion of the Gstrand in the V-set domain forming β-sheet-like hydrogen bonds with the glycerol side chain of the sialic acid. Only few direct protein contacts to the sub-terminal sugars are observed and mediated by the highly variable GG′ linker and CC′ loop. These structural observations in conjunction with surface plasmon resonance binding assays provide mechanistic insights into the linkage-dependent Siglec carbohydrate recognition and suggest that Siglec-5 and other CD33-related Siglec receptors are more promiscuous in sialo-glycan recognition than previously understood.

... ds of scientific knowledge are reflected in the content aspect. Regarding the mode of training research, the emphasis is on reading strategies and activities aimed at fluent comprehension and handling professional and scientific information. Students' scientific conference survey proves the potential of EFL in research activities and sheds the light on the new ways to develop research competence. 26 shs SHS Web of Conferences 1097 E PA R 5 4 The following survey results may be noted as positive ones: the main motive of participation in the conference of the students of all levels of education (those undertaking bachelor's and master's degreeprograms, and additional education program) is their interest to the topic chosen and their attitude to the foreign language they study -80 % of respondents respectively; 70 % of the participants were senior undergraduates, although English is taught for the first-and second-year students doing Bachelor's degree; English and Russian sources were used in almost equal propotion, and the usage of Russian materials means that translation as one of the main types of written communication was implemented; more than 50 % of the participants showed their intention to continue their research on the topic chosen. The number of sources adapted varied between 2 and 3, and it is not enough especially for graduate students. With regard to the composition of the rest of the participants, it was distributed as follows: 20% were first-year students, and only 10 %were students undertaking Master's degree programsfor whom research work is, in fact, the main focus of study.

... copy. The proteomic analysis, multiplex analysis and immunostaining were used to characterize the molecular composition of the CIMVs-MSCs. The transfer of surface proteins from a donor to a recipient cell mediated by CIMVs-MSCs was demonstrated using immunostaining and confocal microscopy. The angiogenic potential of CIMVs-MSCs was evaluated using in vivo approach of subcutaneous implantation of CIMVs-MSCs in mixture with Matrigel matrix. Results: Human CIMVs-MSCs retain parental MSCs content such as growth factors, cytokines, chemokines: EGF, FGF-2, Eotaxin, TGF-α, G-CSF, Flt-3L, GM-CSF, Fractalkine, IFNα2, IFN-γ, GRO, IL-10, MCP-3, IL-12p40, MDC, IL-12p70, IL-15, sCD40L, IL-17A, IL-1RA, IL-1a, IL-9, IL-1b, IL-2, IL-4, IL-5, IL-6, IL-7, IL-8, IP-10, MCP-1, MIP_1a, MIP-1b, TNF-α, TNF-β, VEGF. CIMVs-MSCs also have the expression of surface receptors similar to those in parental human MSCs (CD90+, CD29+, CD44+, CD73+). Additionally, CIMVs-MSCs could transfer membrane receptors to the surfaces of target cells in vitro. Finally, CIMVs-MSCs can induce angiogenesis in vivo after subcutaneous injection into adult rats. Conclusions: Human CIMVs-MSCs have similar content, immunophenotype and angiogenic activity to those of the parental MSCs. Therefore, we believe that human CIMVs-MSCs could be used for cell free therapy of degenerative diseases.
On Assignment of the Upper Bohl Exponent for Linear Time-Invariant Control Systems in a Hilbert Space by State Feedback
2020
Mathematics
We consider a linear continuous-time control system with time-invariant linear bounded operator coefficients in a Hilbert space. The controller in the system has the form of linear state feedback with a time-varying linear bounded gain operator function. We study the problem of arbitrary assignment for the upper Bohl exponent by state feedback control. We prove that if the open-loop system is exactly controllable then one can shift the upper Bohl exponent of the closed-loop system by any

Improving the representation of peptide-like inhibitor and antibiotic molecules in the Protein Data Bank
2014
Biopolymers
With the accumulation of a large number and variety of molecules in the Protein Data Bank (PDB) comes the need on occasion to review and improve their representation. The Worldwide PDB (wwPDB) partners have periodically updated various aspects of structural data representation to improve the integrity and consistency of the archive. The remediation effort described here was focused on improving the representation of peptidelike inhibitor and antibiotic molecules so that they can be easily

... fied and analyzed. Peptide-like inhibi-tors or antibiotics were identified in over 1000 PDB entries, systematically reviewed and represented either as peptides with polymer sequence or as single components. For the majority of the single-component molecules, their peptide-like composition was captured in a new representation, called the subcomponent sequence. A novel concept called "group" was developed for representing complex peptide-like antibiotics and inhibitors that are composed of multiple polymer and nonpolymer components. In addition, a reference dictionary was developed with detailed information about these peptide-like molecules to aid in their annotation, identification and analysis. Based on the experience gained in this remediation, guidelines, procedures, and tools were developed to annotate new depositions containing peptide-like inhibitors and antibiotics accurately and consistently.

Antibiotics
COVID-19 has had a significant impact on health care systems, including drug use. The present study aimed to evaluate the patterns of community supply of antimicrobials from community pharmacies during the COVID-19 pandemic in five cities of Russia. In a cross-sectional study, a random sample of pharmacies reported all episodes of antimicrobials supply during a one-week period. Patterns of supply (age and gender of customer, drug name and formulation, prescription availability, indication,

... were analyzed. Altogether, 71 pharmacies took part in the study and 5270 encounters were recorded. In total, 4.2% of visits resulted in supply of more than one antimicrobial agent and 5.2% were for parenteral formulations. The rate of prescription-based purchase in participated cities varied from 40.5 to 99.1%. Systemic antibiotics and antivirals accounted for the majority of supplies (60.5 and 26.3%, respectively). Upper respiratory tract infections were reported as the indication for antimicrobials usage in 36.9% of cases, followed by skin and soft tissue infections (12.1%) and urinary tract infections (8.7%); COVID-19 accounted for 8.4% of all supplies. Amoxicillin with clavulanic acid, azithromycin and amoxicillin were indicated as the top three antimicrobials purchased for upper respiratory tract infections, and azithromycin, umifenovir and levofloxacin were the top three for COVID-19. In general, a high rate of drugs dispensing without prescription was revealed. Antibiotics for systemic use remained the most common antimicrobials, whereas presumably viral upper respiratory tract infections were the main reason for their purchase. COVID-19 infection itself was responsible for a small proportion of the supply of antimicrobial agents, but systemic antibiotics accounted for more than a half of supplies.
