Comparison of tissue effects in rabbit muscle of surgical dissection devices

While some energy-based surgical dissection and coagulation modalities may offer excellent cutting and coagulation abilities, the impact on healing may differ among devices. We compared the tissue effects of three of these modalities with those of the standard surgical scalpel in rabbit muscle at 24 h and 14 days after surgery by evaluating radiographic and histological data. Linear incisions were made with each device in the dorsal lumbar musculature of rabbits using monopolar electrocautery

... cut mode (MPE-Cut) and coagulation mode (MPE-Coag), a ferromagnetic induction loop (FMI), and a traditional scalpel. Magnetic resonance imaging scans and histological sampling were done at 24 h and 14 days. Subjective cutting and coagulation characteristics for each device were also recorded during surgery. The scalpel and FMI appeared to cause the least tissue damage adjacent to the incisions in rabbit dorsal lumbar musculature. The scalpel showed the best healing, while the FMI and MPE-Cut demonstrated good healing. The MPE-Coag showed the worst tissue healing. The scalpel, FMI, and MPE-Cut all exhibited favorable subjective characteristics during surgery. It appears that the FMI may be a better choice for surgical dissection and coagulation in muscle tissue than the MPE coagulation mode because it shows less tissue damage and offers better tissue healing.

Treating ACL injuries in prepubescent patients requires balancing the risk of chondral and meniscal injuries associated with delaying treatment against the risk of growth disturbance from early surgical reconstruction. Multiple physeal respecting techniques have been described to address this vulnerable population; however, none restore the native ACL attachments while keeping the graft and fixation entirely in the epiphysis.

Expression of the Rho-GEF Pbl/ECT2 is regulated by the UBE3A E3 ubiquitin ligase

We applied genetic tools available in Drosophila to identify candidate substrates of the UBE3A ubiquitin ligase, the gene responsible for Angelman syndrome (AS). Human UBE3A was expressed in Drosophila heads to identify proteins differentially regulated in UBE3A-expressing versus wild-type extracts. Using two-dimensional gel and MALDI-TOF analysis, we detected 20 proteins that were differentially regulated by over-expression of human UBE3A in Drosophila heads. One protein responsive to UBE3A

... the Rho-GEF pebble (pbl). Here, we present three lines of evidence suggesting that UBE3A regulates Pbl. First, we show genetic evidence that UBE3A and the Drosophila de-ubiquitinase fat facets (faf) exert opposing effects on Pbl function. Secondly, we find that both Pbl and ECT2, the mammalian orthologue of Pbl called epithelial cell transforming sequence 2 oncogene, physically interact with their respective ubiquitin E3 ligases. Finally, we show that Ect2 expression is regulated by Ube3a in mouse neurons as the pattern of Ect2 expression is dramatically altered in the hippocampus and cerebellum of Ube3a null mice. These results suggest that an orthologous UBE3A post-translational regulatory pathway regulates neuronal outgrowth in the mammalian brain and that dysregulation of this pathway may result in neurological phenotypes including AS and possibly other autism spectrum disorders.

The interaction of dislocations with and near grain boundaries plays an important role in the strain response of metals. Whether a dislocation is blocked or transmitted at a grain boundary is dependent on a number of inter-related factors including the alignment of slip systems, the inclination of the interface, and the nature of the defect transition states within the interface itself. In this presentation we consider the morphology and character of dislocations interacting with {111} and

... twin boundary facets in Ni. Our work is motivated in part by interest in explaining the influence of twin boundaries on the fracture behavior of metals exposed to hydrogen environments. For instance, Bechtle et al. have reported a reduced susceptibility to intergranular hydrogen embrittlement in Ni processed to have a high fraction of Σ3 twins and related Σ3 n special boundaries [1]. However, analyses have also shown that although they are resistant to crack propagation, twins can still be sites of preferential crack initiation during loading in the presence of hydrogen [2], raising questions regarding the precise mechanisms of strain localization at such interfaces and how these are changed with exposure to hydrogen. To address these questions, we are investigating dislocation/twin interactions using Diffraction Contrast Scanning Transmission Electron Microscopy (DC-STEM) [3] . Figure 1(a) shows a dark-field DC-STEM image of dislocations in the vicinity of a Σ3 twin in Ni, which has formed {111} and {112} facets. This material was precharged with 4000 appm H 2 and then deformed at room temperature to 3.5% strain. The higher magnification image in Figure 1 (b) shows a network of grain boundary dislocations that have formed on one of the twin boundary facets. We are also conducting tilt series so that we can reconstruct the complex three-dimensional topology of the dislocations both within and in the vicinity of the twin boundaries. Figure 2 shows one such example from a series tilting about a Σ3 {112} facet. As we will discuss, the configuration of these defect networks can be understood in terms of the underpinning interfacial bicrystallography, which predicts the presence of Burgers vectors of type (1/6)<112> and (1/3)<111>.
