Effective handling of location-based data is important to emergency response management (ERM). Expert team members co-located around maps typically discuss events while drawing freeform areas or while using physical placeholders representing incidents. Key ERM functions are filtering data, selecting information recipients, searching datasets, drawing time-dependent freeform areas, and zooming in on one region while leaving others unchanged. more &raquo; ... ufficient; intuitive graspable solutions, such as tangible user interfaces (TUIs), may be better suited for ERM. We present CoTracker, a tangible tabletop system with expected potential for ERM teamwork. On an interactive map expert team members can discuss an operational picture using TUIs like bricks, frames, and pens. With the participation of domain experts for cognitive walkthrough studies, we examined how generic and specialized TUIs can support ERM-related functions. We present some insights into the design of ERM-focused tangible tabletops.

High-Temperature Short-Time Treatment of Human Milk for Bacterial Count Reduction

Human milk (HM) for preterm infants will often be pasteurized for cytomegalovirus (CMV) inactivation and reduction of its bacterial count. High-temperature short-time (HTST) treatment compared to standard Holder pasteurization (HoP) reduces the impact of heat treatment on bioactive HM proteins while effectively inactivating CMV. No data are available for the efficacy of bacterial count reduction using HTST treatments that are available for clinical use. Objective: To test the antiviral and
more &raquo; ... acterial efficacy of HTST treatment protocols in HM using a modified HTST treatment device compared to standard HoP. Methods: Holder pasteurized 95 mL HM samples were inoculated with Staphylococcus aureus (ATCC 6538), Enterococcus faecalis (ATCC 29212), Pseudomonas aeruginosa (ATCC 27853), Serratia marcescens (Smarc 00697), two different strains of Klebsiella pneumoniae (ATCC 700603 and Kpn 01605) or spiked with 2 × 105 50% tissue culture infective dose of CMV (AD169) and subsequently subjected to HoP (62.5°C/30 min) or HTST treatment (62°C/5 s, 62°C/15 s, 72°C/5 s, 72°C/15 s, 87°C/2 s, and 87°C/5 s). Bacterial count was determined after treated HM was cultured for 24 h. CMV infectivity was determined by the number of specific CMV immediate early antigen stained nuclei after inoculating human fibroblasts with appropriately prepared HM samples. Results: Holder pasteurized samples revealed no growth after 24 h incubation. Viable bacterial cultures were retrieved from all tested strains after HTST treatment with the default HTST protocol (62°C/5 s) that is available for clinical use. Using other time-temperature combinations, growth rates of S. aureus, E. faecalis, P. aeruginosa, K. pneumoniae, K. pneumonia, and S. marcescens were depending on treatment time, treatment temperature, bacterial genera and strain. Only after treatment temperatures above 72°C no bacterial growth was observed. CMV was inactivated by any tested time-temperature combination. Conclusions: HTST treatment inactivates CMV in 95 mL HM samples but is less effective than HoP in bacterial count reduction at a time-temperature combination of 62°C/5 s. For a reliable bacterial count reduction HTST treatment at 87°C was required in this study.

Eps15-homology domain containing protein 2 (EHD2) is a dynamin-related ATPase located at the neck of caveolae, but its physiological function has remained unclear. Here, we found that global genetic ablation of EHD2 in mice led to increased fat accumulation. This organismic phenotype was paralleled at the cellular level by increased lipid uptake via a caveolae-, dynamin- and CD36-dependent pathway, an elevated number of detached caveolae and higher caveolar mobility. Furthermore, EHD2
more &raquo; ... itself was down-regulated in the visceral fat of two obese mouse models and obese patients. Our data suggest that EHD2 controls a cell-autonomous, caveolae-dependent lipid uptake pathway and suggest that low EHD2 expression levels are linked to obesity.

Staphylococcal alpha-toxin kills human keratinocytes by permeabilizing the plasma membrane for monovalent ions

Incubation of human keratinocytes with nanomolar concentrations of Staphylococcus aureus alpha-toxin leads to irreversible depletion of cellular ATP. The toxin forms hexamers in the target cell membranes, and rapid transmembrane flux of K+, Na+, and 86Rb+ is observed. Unexpectedly, pores formed in keratinocytes through application of low but lethal doses of alpha-toxin appeared to be considerably smaller than those formed in erythrocyte membranes. They permitted neither rapid influx of Ca2+ or
more &raquo; ... ropidium iodide, nor efflux of carboxyfluorescein. Larger pores allowing flux of all three markers did form when the toxin was applied at high concentrations. Flux of monovalent ions and reduction in cellular ATP levels evoked by low toxin doses correlated temporally with a fall in oxygen consumption, which was interpreted to reflect breakdown of mitochondrial respiration. The lethal event could not be thwarted by manipulating the extracellular K+ or Ca2+ concentrations. Realization that alpha-toxin may form very small pores in nucleated cells is important for future research on cellular toxin effects and membrane repair processes.

Face morph detection for unknown morphing algorithms and image sources: a multi-scale block local binary pattern fusion approach

The vulnerability of face recognition systems against so-called morphing attacks has been revealed in the past years. Recently, different kinds of morphing attack detection approaches have been proposed. However, the vast majority of published results has been obtained from rather constrained experimental setups. In particular, most investigations do not consider variations in morphing techniques, image sources, and image post-processing. Hence, reported performance rates can not be maintained
more &raquo; ... n realistic scenarios, as the NIST FRVT MORPH performance evaluation showed. In this work, existing algorithms are benchmarked on a new, more realistic database. This database consists of two different data sets, from which morphs were created using four different morphing algorithms. In addition, the database contains four different post-processings (including print-scan transformation and JPEG2000 compression). Further, a new morphing attack detection method based on a fusion of different configurations of Multi-scale Block Local Binary Patterns (MB-LBP) on an image divided into multiple cells is presented. The proposed score-level fusion of a maximum number of 18 different configurations is shown to significantly improve the robustness of the resulting morphing attack detection scheme, yielding an average performance between 2.26% and 8.52% in terms of Detection Equal Error Rate (D-EER), depending on the applied post-processing.

Characterization of circulating plasma proteins in dairy cows with cytological endometritis

Early diagnosis of endometritis in dairy cattle is currently requires invasive techniques and specialist expertise. The goal of this study is to utilize a gel-free mass-spectrometry based proteomics approach to compare the plasma proteome of dairy cattle with cytological endometritis to those without. Blood samples were collected from cows (N = 112) seven days postpartum (DPP). Plasma samples from a cohort of 20 animals with cytological endometritis (n = 10) and without (n = 10) as classified
more &raquo; ... DPP were selected for proteomic analysis. Differential abundances of proteins between the two animal groups were determined using both fold change (≥1.5 fold change) and statistical significance threshold (p < .05). A total of 181 non-redundant proteins were quantified, and 25 proteins were found with differential abundance. These include 4 binding protein alpha and mannose binding lectin 2 involved in immune responses. Differentially abundant proteins between the animals were then processed using PANTHER for gene ontology. Gene ontology included associations with innate immune processes, acute phase responses and immune regulation. A potential marker for disease identified here is the "uncharacterized protein G5E513," a protein previously defined by RNA-transcripts. These proteins may form the basis for endometritis prognosis, the development of which is proceeded by systemic changes in immune function. SIGNIFICANCE: Endometritis is a costly reproductive disease of lactating dairy cows that warrants timely diagnosis. We utilized a gel-free mass-spectrometry based proteomics approach to compare the plasma proteome of dairy cattle with cytological endometritis to those without, for the characterization of changes in the proteomic profile associated with uterine disease postpartum. Furthermore, we compared the plasma proteome of healthy and affected cows in the same physiological status of production to better understand the relationship between changes in expression of circulating proteins and to unravel essential biological mechanisms involved in bovine cytological endometritis.
