Expression of Proteus mirabilis polyphosphate kinase and preparation of its polyclonal antibodies

PPK fusion protein expressed by E. coli was purified successfully using Ni-affinity chromatography. ELISA result demonstrated that the harvested rabbit anti-sera against PPK had a high titer of 1:512 000, and Western blotting showed a good specificity of the antibody, which can be used further study of the role of PPK in the pathogenesis of Proteus mirabilis infection.

Status of Human Papillomavirus Vaccination and Knowledge,Attitudes, and Influencing Factors towards Human Papillomavirus and Its Vaccines among University Students in Western China

Objective To investigate the knowledge about human papillomavirus(HPV)and its vaccines,HPV vaccination status,and vaccination willingness among the freshmen majoring in liberal arts,sciences,and medicine in western China,so as to provide evidence-based suggestions for health education among university students and to increase their vaccination. Methods By convenience sampling,stratified sampling,and cluster sampling,an online questionnaire survey was conducted among freshmen in a comprehensive
more » ... niversity in Chengdu.The differences among the groups were compared by χ2 test,and non-conditional logistic regression was performed to analyze the influencing factors of willingness to vaccination. Results A total of 881 participants were included in this study,including 419 males and 462 females.The vaccination rate of females was 3.5%(16/462).Of all the university students,59.1%,67.5%,and 56.3% had heard of HPV,HPV-related diseases,and HPV vaccines,respectively,and only 10.4%,10.6%,and 27.8% knew "HPV infection usually does not cause symptoms","HPV infection is usually transient and mostly spontaneous regressive",and "HPV infection is very common".Females and medical students had more related knowledge than males and non-medical students(P=0.035,P=0.021).Of all the university students,65.7% were willing to be vaccinated against HPV.The high cost of vaccination(50.8%),worry about the side effects adverse reaction(46.3%),and lack of sexual life(43.4%)were the main reasons for the reluctance to be vaccinated.Gender,education background of parents,history of self-financed vaccination,attitude towards premarital sexual behavior,history of sexual behavior,and knowledge of HPV were the influencing factors of vaccination willingness. Conclusions The freshmen had a low rate of HPV vaccination and limited detailed knowledge about HPV and its vaccines.Health education campaign is urgently called for among university students regarding HPV infection as well as the safety,effectiveness,and side effects of HPV vaccines.More attention should be paid to the health education of male and non-medical students.

Verification of a sporadic Alzheimer disease model in SORL1 gene knockout mice

To compare the behavioral and pathological features of SORL1 gene knockout mice with those of normal mice and APP/PSE1 mice to verify the feasibility of using SORL1 knockout mice as a model of sporadic Alzheimer disease. more » ... rris water maze test to assess their learning and memory abilities with positioning navigation and space exploration experiments. The expression of APP and Aβ in the brain of the mice was detected using immunohistochemistry and Western blotting, respectively. DNA sequencing showed CAAT deletion in SORL1 gene in two chromosomes of SORL1-/- mice, and the control mice had intact SORL1 gene without the deletion; Western blotting did not detect the expression of the SORL1 in the brain of SORL1-/- mice. Morris water maze test showed that in positioning navigation experiment, the average avoidance latency was similar between SORL1-/- mice and APP/PSE1 mice (P>0.05) but increased significantly in both mice as compared with the control group (P<0.05); similar results were obtained in the space exploration experiment. Immunohistochemistry and Western blotting revealed significantly increased APP and Aβ expression in the brain tissue of both SORL1-/- mice and APP/PSE1 mice compared with the control mice without significant differences between the two transgenic mice. SORL1-/- mice exhibit similar behavioral and pathological changes with APP/PSE1 mice and can be used as a model of sporadic Alzheimer's disease.

To establish the diagnostic criteria for polypoidal choroidal vasculopathy (PCV) based on spectral-domain optical coherence tomography (SD OCT) by evaluating the sensitivity and specificity of SD OCT in differentiating PCV from wet age-related macular degeneration (wAMD). more » ... on findings by fundus photography, fluorescein angiography (FFA) and indocyanine green angiography (ICGA). Among the 6 features of SD OCT, namely a sharp RPED peak, double-layer sign, multiple RPED, an RPED notch, a hyporeflective lumen representing polyps, and hyperreflective intraretinal hard exudates, findings of the first two features and at least one of the other features sufficed the diagnosis of PCV; in the absence of the first two features, the diagnosis of PCV was also made when at least 3 of the other features were present simultaneously. The sensitivity and specificity of SD OCT-based diagnosis were estimated by comparison with the gold standard ICGA-based diagnosis. In the 25 eyes with an established diagnosis of PCV, 23 eyes (92.0%) met the diagnostic criteria based on SD OCT findings; in the 38 eyes with the diagnosis of wAMD, only 4 eyes (10.5%) met the criteria. The sensitivity and specificity of SD OCT-based diagnosis of PCV was 92.0% and 89.5%, respectively. s We established the diagnostic criteria for PCV based on SD OCT findings with a high sensitivity and specificity. SD OCT shows a strong capacity for differentiating PCV from wAMD.

The latest guide is "New Coronary Virus-Infected Pneumonia Diagnosis and Treatment Plan (Seventh Trial Version)"(short for Seventh Version of Diagnosis and Treatment Plan). But the use of antiviral drugs is still on trial and no rigorous clinical trials data is available. Hot anti-SARS-CoV-2 drugs include interferon α, ribavirin, lopinavir/ritonavir, chloroquine phosphate, abidol, as well as hydroxychloroquine sulfate and remdesivir. But the later 2 drugs aren't mentioned in the Seventh Version of Diagnosis and Treatment Plan.

Cervical heterotopic pregnancy: A case report

Heterotopic pregnancies are rare and difficult to be diagnosed early. A patient with combined intrauterine pregnancy and cervical pregnancy was admitted in Qingdao Municipal Hospital in 2019. The patient complained of abnormal vaginal bleeding after menopause and was misdiagnosed as simple intrauterine pregnancy. She underwent artificial abortion and suffered intraoperative hemorrhage. A probability model for analyzing speckles in intravascular ultrasound images to facilitate image segmentation

Ultrasonic image speckles result from the interference of the reflected signals by the scatters in the detected tissue. The physical characteristics of the speckles are closely correlated with the structures of the biological tissues, and the probability distribution of these speckles differs across different tissues. more » ... , soft plaque and normal vascular regions on IVUS images. Using KS test, KL divergence and correlation coefficient analysis, we found that the probability distributions of the speckles generated by calcified plaques and normal blood vessels were better described by the Gaussian mixture model, while the speckles caused by soft plaques were described better by the Gamma mixture model. Based on this finding, we propose a probability mixture model combining neighborhood information for plaque segmentation on IVUS images. Compared with the existing probabilistic mixture model, the segmentation accuracy was greatly improved with a reduced noise.

Protective effect of butylphthalide in a cell model of Alzheimer's disease induced by Aβ25-35 in Neuro 2a cells

To study the protective effect of butylphthalide in a cell model of Alzheimer's disease(AD) induced by Aβ25-35 in Neuro 2a (N2a) cells. N2a cells were divided into AD group, butylphthalide (NBP) group and control group. AD cell model was established by adding 20 µmol/L Aβ25-35 to cultured N2a cells. more » ... ected by AnnexinV-FITC flow cytometry, and the cell morphological changes were observed under inverted phase contrast microscope. The expression of TNF-α and IL-1β mRNA were determined by qRT-PCR. Compared with those in the control group, the number of adherent cells was significantly decreased, neurite structures were reduced, and the cell viability was decreased, while the apoptotic rate and expressions of TNF-α and IL-1β mRNA were increased in AD group (P<0.05). Compared with that in AD group, the number of adherent cells was increased in NBP group and the cell morphology was similar to the normal control cells. The cell viability of N2a cells was increased in NBP group with decreased apoptotic rate and expression of TNF-αand IL-1β mRNA (P<0.05). Butylphthalide can protect against AD in the cell model induced by Aβ25-35 possibly by inhibiting the expression of inflammatory cytokines.

We have developed genetically encoded fluorescent sensors for reduced nicotinamide adenine dinucleotide (NADH), which manifest a large change in fluorescence upon NADH binding. We demonstrate the utility of these sensors in mammalian cells by monitoring the dynamic changes in NADH levels in subcellular organelles as affected by NADH transport, glucose metabolism, electron transport chain function, and redox environment, and we demonstrate the temporal separation of changes in mitochondrial and

These results support the view that cytosolic NADH is sensitive to environmental changes, while mitochondria have a strong tendency to maintain physiological NADH homeostasis. These sensors provide a very good alternative to existing techniques that measure endogenous fluorescence of intracellular NAD(P)H and, owing to their superior sensitivity and specificity, allow for the selective monitoring of total cellular and compartmental responses of this essential cofactor.

Inhibiting HSP70 expression enhances cisplatin sensitivity of cervical cancer cells

To investigate the relationship between sensitivity to cisplatin (DDP) and the expression of HSP70 in cervical cancer cells in vitro. more » ... essions of HSP70, Bcl-2, Bax and caspase-3 were measured with Western blotting. A nude mouse model bearing Hela229 cell xenograft was used to evaluate the effect of DDP and PFT-µ on tumor growth. Hela229 cells expressed a higher level of HSP70 than normal cervical cells. The combined use of PFT-µ significantly enhanced the inhibitory effect of DDP (P<0.01) and increased the cell apoptosis in Hela229 cells. JC-1 staining demonstrated that DDP combined with PFT-µ more obviously reduced mitochondrial membrane potential. DDP combined with PFT-µ more strongly lowered Bcl-2 expression and increased the expressions of casepase-3 and Bax than DDP alone. In the nude mouse model, PFT-µ significantly enhanced DDP sensitivity of Hela229 cell xenografts (P<0.01). Inhibition of HSP70 expression can enhance the sensitivity of cervical cancer cell to DDP both in vivo and in vitro possibly by promoting cell apoptosis, suggesting the potential of HSP70 as a new target for gene therapy of cervical cancer.
