The HPLC-UV analysis revealed the presence of depsidones, 3-hydroxyphysodic, 4-O-methyl physodic acid, physodic and physodalic acid together with two depsides, atranorin and chloroatranorin. GC-FID and GC-MS analyses enabled the identification of atranol, chloroatranol, atraric acid, olivetol, olivetonide and 3-hydroxyolivetonide as the main components. The results of present study show that H. tubulosa acetone extract is a promising candidate for in vivo experiments considering antioxidant activity.

Thiobarbituric acidreacting substances (TBARS), SH groups (SH), total glutathione (GSH), glutathione peroxidase activity (GPx), superoxide dismutase activities (tSOD, MnSOD, CuZnSOD) and nitrite/nitrate concentration (NO 2 +NO 3 ) were assessed in plasma and brain structures (whole encephalitic mass-WEM and brainstem-BS). Results: Obtained results showed that AGM treatment successfully attenuated severe clinical deficits in EAE. Applications of AGM in EAE rats induced normalized TBARS, SH, GSH, GPx and NO in WEM. In BS, AGM expressed less prominent effects, inducing normalized TBARS, GPx and NO, but no effect on SH and GSH. In both brain structures, tSOD activity lowered and normalized at the peak and in the remission phase of the disease, post-AGM treatment. The effect of AGM on the MnSOD in EAE was expressed in WEM/BS only in the remissive phase as a reduced activity. Conclusion: Milder clinical form of developed EAE in rats indicates promising therapeutic effect of AGM in MS. The activated antioxidant system and suppressed oxidative/ nitrosative stress development may denote a successful blockade of neuroinflammation initiated by EAE immunization. The study implies the capability of AGM to attenuate oxidative/ nitrosative damage in the peak of EAE by modulating antioxidative defense capacity during the time-course of the disease. Thus, AGM may be considered as an agent with a beneficial effect on neuroinflammation in EAE. Abstrakt Uvod: Eksperimentalni autoimunski encefalomijelitis (EAE) je model multiple skleroze (MS), u kome su ispitivani neuroprotektivni efekti agmatina (AGM), poznatog primarnog amina koji se dobija dekarboksilacijom L-arginina. Metode: Dark Aguti pacovi su 4 podeljeni u grupe: kontrola (C), kompletni Frojdov adjuvans (CFA), EAE pacovi dekapitovani 13 dana (EAE13) i 20 dana (EAE20) nakon imunizacije, EAE životinje sa tri (EAE+AGM13) i 10 (EAE+AGM20) doza AGM i zdrave životinje sa tri/10 doza AGM (AGM). Reaktivne supstance koje reaguju sa tiobarbiturnom kiselinom (TBARS), SH grupe (SH), koncentracija ukupnog glutationa (GSH), aktivnost glutation peroksidaze (GPx), aktivnost superoksid dizmutaza (tSOD, MnSOD, CuZnSOD) i koncentracija nitrita/nitrata (NO 2 +NO 3 ) su određivani u plazmi i moždanim strukturama (kompletna encefalitična masa-WEM i produžena moždina-BS). Rezultati: Dobijeni rezultati pokazuju da tretman sa AGM uspešno smanjuje teški klinički deficit kod EAE. Aplikacija AGM kod EAE pacova normalizuje TBARS, SH, GSH, GPx i NO u WEM. U BS, AGM dovodi do manje izraženih efekata normalizacijom TBARS, GPx i NO, ali nema efekata na SH grupe i GSH. U obe moždane strukture, tSOD je smanjena i normalizovana u piku i remisiji bolesti nakon tretmana sa AGM. Efekat AGM na MnSOD kod EAE je izražen u WEM/BS samo u toku remisije bolesti kao redukcija aktivnosti enzima. Zaključak: Blaža forma razvijenog EAE pokazuje izraženi terapijski efekat AGM kod MS. Aktivirani antioksidativni sistem i supresija razvoja oksidativnog/ nitrozativnog stresa mogu označiti uspešnu blokadu neuroinflamacije indukovanu EAE imunizacijom. Studija pokazuje sposobnost AGM da ublaži oksidativno/ nitrozativno oštećenje u piku EAE modulacijom kapaciteta antioksidativne odbrane u toku trajanja bolesti. U tom smislu, AGM se može smatrati agensom sa antioksidativnim pristupom za lečenje i prevenciju neuroinflamacije u EAE.

The second group was treated with both chlorpromazine (at a single dose of 38.7 mg/kg b.w.) and agmatine (at a single dose of 75 mg/kg b.w.). Agmatine was applied immediately after the chlorpromazine. The control group was treated with 0.9% saline solution in the same manner. Rats were sacrificed by decapitation 24 h after the treatment and biochemical and immunohistochemical examinations were performed. Analysis of data showed that treatment with agmatine significantly attenuated the oxidative stress indicators as evidenced by lowering malonylaldehyde concentrations in the liver and in plasma while not affecting liver concentrations of total glutathione and plasma concentration of sulfhydryl groups. Additionally, histological evaluation revealed the improvement of liver damage in this respect. The presented data indicated that intraperitoneally administered agmatine protects against chlorpromazine-induced liver disease in rats.

... in lymphocyte physiology and found that NUPR1 might be involved in the initiation of their proliferation. Lymphocytes were isolated from the cervical lymph nodes of C57BL/6 mice. NUPR1 expression subsided 24 h after the induction of proliferation by a mitogen. Also, stressful conditions after cell isolation led to increased NUPR1 mRNA and protein expression in vitro that coincided with cell apoptosis. Similarly, apoptosis induction by staurosporine, a broadrange protein kinase inhibitor, led to increased NUPR1 expression. In addition, NUPR1 inhibition by small-interfering RNA prevented the staurosporine-induced apoptosis (judging from decreased caspase activity) in the whole cell population of cervical lymph nodes. However, NUPR1 absence was irrelevant to the induction of apoptosis in CD3 + T lymphocytes, suggesting that NUPR1 is probably a mediator of apoptosis in other immune cell populations within the lymph node, such as B lymphocytes. In conclusion, our results suggest that NUPR1 is important for the initiation of lymphocyte cell division and for the apoptotic process of non-T cells during stressful conditions.

... regnancy and infancy, increased amounts of folate are required. Folate has the roles in the prevention of neural tube defects, an important cause of infant mortality and disability. The periconceptional folic acid intake reduces the risk of neural tube defects. Arginase, as part of the urea cycle, detoxifies human body from ammonia. Its activity is important in the production of ornithine, subsequently leading to the production of polyamines (spermine and spermidine), glutamate or proline, and nitric oxide. Arginase is normally present in mother's milk. In the present study, the effect of folic acid supplementation during pregnancy on the dynamics of arginase activity and malondialdehyde concentration in human colostrum and mature milk was studied. The obtained results suggest that in breastfeeding mothers' colostrum and mature human milk samples, supplemented with folic acid during pregnancy (400µg/daily periconceptionaly), the arginase activity increases compared with the enzyme activity in milk samples taken from mothers who did not take folic acid. At the same time, the supplementation with folic acid causes malondialdehyde decrease in colostrum and mature milk.

To analyze the frequency and intensity of pain and its association with different characteristics of patients with myotonic dystrophy type 1 (DM1) and type 2 (DM2), 52 adult genetically confirmed DM1 and 44 DM2 patients completed the Brief Pain Inventory questionnaire (BPI). Average pain intensity correlated with Individualized Neuromuscular Quality of Life (INQoL) total score in both DM1 (rho = +0.30, p < 0.05) and DM2 patients (rho = +0.61, p < 0.01). In conclusion, the majority of DM1 and DM2 patients have mild to moderate pain. Our results open new opportunities for behavioral and cognitive interventions.
