Due to manufacturing flaws or cyclic loading, cracks frequently appear in a rotating spindle system. These cracks markedly affect the dynamic characteristics in higher modes of the rotating machinery. For faster rotational speeds, especially for super-high-speed cutting, a spindle with magnetic bearings is necessary. A Euler-Bernoulli beam of circular cross section is used to approximate the spindle and the Hamilton principle is employed to derive the equation of motion for the spindle system. Decreased PEDF Expression Promotes Adipogenic Differentiation through the Up-Regulation of CD36

Adipogenesis is a tightly regulated cellular process that involves the action of multiple signaling pathways. Characterization of regulators that are associated with adipose development is crucial to understanding the mechanisms underlying obesity and other metabolic disorders. Pigment epithelium-derived factor (PEDF) is a secreted glycoprotein that was first described as a neurotrophic factor. The role of PEDF in lipid metabolism was established when adipose triglyceride lipase (ATGL), a major
more &raquo; ... triglyceride hydrolase, was characterized as its binding partner. In this study, we investigated the downstream effects of PEDF on adipogenic differentiation using rat adipose-derived stem cells (AdSCs) and the mouse pre-adipocyte cell line 3T3-L1. Knocking down PEDF in differentiating cells resulted in elevated levels of ATGL and CD36, as well as other adipogenic markers, with a concomitant increase in adipocyte number. CD36, a scavenger receptor for a variety of ligands, regulated proliferation and lipogenic gene expression during adipogenesis. The CD36 increase due to PEDF down-regulation might be a result of elevated PPARγ. We further demonstrated that PEDF expression was regulated by dexamethasone, a synthetic glucocorticoid that is widely used for adipogenesis at the transcriptional level. Taken together, our findings highlight that PEDF negatively regulates adipogenesis through the regulation of various signaling intermediates, and it may play a crucial role in lipid metabolic disorders.

In high temperature applications, the oxidation of polymer composites changes their microstructures and degrades their mechanical properties. Both these effects detrimentally affect the service life. Oxidation experiments are time-consuming and cannot be conducted in a laboratory for all layouts of a composite. For this reason, the weight loss of a composite due to oxidation is predicted by the first-order approximation rule of mixtures. Theoretically, the shielding of fibers should slow down
more &raquo; ... e oxidation of a composite than that of a neat resin. However interestingly, the predicted value of oxidation is far lower than that obtained experimentally. In the literature, some researchers have proposed the fibermatrix interface model to explain the discrepancy while others have believed it is related to the preparation of the surface. This work presents a surface roughness model to clarify the relationship between the high-temperature oxidation and the surface roughness. Predictions concerning high-temperature oxidation are favorably supported by experimental data.

Decreased PEDF Promotes Hepatic Fatty Acid Uptake and Lipid Droplet Formation in the Pathogenesis of NAFLD

Non-alcoholic fatty liver disease (NAFLD), the leading cause of chronic liver diseases worldwide, ranges from simple steatosis to steatohepatitis, with the risk for progressive fibrosis or even cirrhosis. While simple steatosis is a relatively benign condition, the buildup of toxic lipid metabolites can induce chronic inflammation, ultimately triggering disease progression. more &raquo; ... promotes lipolysis through binding to adipose triglyceride lipase (ATGL), a key enzyme for triglyceride breakdown. In the current study, we aimed to delineate how changes in PEDF expression affect hepatic lipid accumulation. Our data revealed that hepatic PEDF was downregulated in a mouse NAFLD model. We further showed that decreased PEDF levels in hepatocytes in vitro resulted in elevated fatty acid uptake and lipid droplet formation, with concomitant upregulation of fatty acid transport proteins CD36 and fatty acid binding protein 1 (FABP1). RNA sequencing analysis of PEDF knocked down hepatocytes revealed an alteration in gene expression profile toward lipid accumulation. Additionally, decreased PEDF promotes mobilization of fatty acids, an observation distinct from blocking ATGL activity. Taken together, our data suggest that hepatic PEDF downregulation causes molecular changes that favor triglyceride accumulation, which may further lead to NAFLD progression.

A 915 MHz printed radio-frequency-identification (RFID) antennas with the characteristics of good gain and omnidirectional beam wave is constructed and evaluated in this study. The objective is to find out their best reading rates for providing effective wireless communications among RFID antenna during the library book inventory process. And an optimal library inventory system which is based on electromagnetic identification (EMID) technology is proposed, which is constructed to find the
more &raquo; ... l tag location for a book, test the tag readability for bookshelves, and connect a couple of multi-layer bookshelves with multiplexers and updat the tag reading status in the database of the computer terminal. The fabricated antenna reader and the proposed system are embedded into different locations of bookshelves and tested at the library of Cheng Shiu University in Taiwan. According to the experimental results, the designed prototype of the antenna reader has the characteristics of the directional radiation pattern, good gain, simple shape, low cost and is easy to be integrated into the bookshelf. And the designed library inventory system can authenticate the location of a book automatically . They can benefit administrating librarians with the capabilities of decreasing the library inventory processing time and reducing the possibility of the books being misplaced.

Double autophagy modulators reduce 2-deoxyglucose uptake in sarcoma patients

Rationale: According to the metabolic symbiosis model, cancer stromal fibroblasts could be hijacked by surrounding cancer cells into a state of autophagy with aerobic glycolysis to help provide recycled nutrients. The purpose of this study was to investigate whether combined treatment with the autophagy inhibitor: hydroxychloroquine (HCQ) and the autophagy inducer: sirolimus (rapamycin, Rapa) would reduce glucose utilization in sarcoma patients. Methods: Ten sarcoma patients who failed
more &raquo; ... e treatment were enrolled in this study. They were treated with 1 mg of Rapa and 200 mg of HCQ twice daily for two weeks. The standardized uptake values (SUV) from pretreatment and posttreatment [18F]-fluorodeoxyglucose positron emission tomography (FDG PET) scans were reviewed, and changes from the baseline SUVmax were evaluated. Results: Based on FDG PET response criteria, six patients had a partial response; three had stable disease, and one had progressive disease. Nevertheless, none of them showed a reduction in tumor volume. The mean SUVmax reduction in the 34 lesions evaluated was − 19.6% (95% CI = −30.1% to −9.1%), while the mean volume change was +16.4% (95% CI = +5.8% to + 27%). Only grade 1 toxicities were observed. Elevated serum levels of lactate dehydrogenase were detected after treatment in most metabolic responders. Conclusions: The results of reduced SUVmax without tumor volume reduction after two weeks of Rapa and HCQ treatment may indicate that non-proliferative glycolysis occurred mainly in the cancer associated fibroblast compartment, and decreased glycolytic activity was evident from Rapa + HCQ double autophagy modulator treatment.

Adipose-derived mesenchymal stem cells (ASCs) are readily available multipotent mesenchymal progenitor cells and have become an attractive therapeutic tool for regenerative medicine. We herein investigated the mechanistic role of how miR-27b modulated regenerative capacities of ASCs. Intravenous administration of miR-27b-transfected ASCs (ASCs-miR-27b) was conducted after 70% partial hepatectomy (PH). We showed that the nature of ASCs-miR-27b to inhibit hepatic stellate cell activation was dependent upon peroxisome proliferator-activated receptor gamma coactivator-1 alpha (PGC-1α) in vitro. Moreover, expression of miR-27b in ASCs induced heme oxygenase-1 (HO-1), resulting in increased production of ATP, protective cytokines/growth factors, and genes involved in mitochondrial biogenesis in a PGC-1α-dependent manner. RNA sequencing (RNA-Seq) analysis revealed drastic transcriptional changes in livers treated with ASCs-miR-27b after PH. The differentially expressed genes classified into "regeneration," "fibrosis," and "mitochondrial biogenesis" clusters were mainly mitochondrial. The potential biological context reflecting the effects of PGC-1α by ASCs-miR-27b treatment was also observed by the subnetwork analysis with HO-1 and PGC-1α being the top-ranked regulatory genes. We demonstrate autologous ASCs-miR-27b enhances liver regeneration and, importantly, preserves hepatic function through paracrine actions which offers a viable therapeutic option to facilitate rapid recovery after liver injury.

A compact low insertion loss bandpass filter based on meandered self-coupled ring resonator

The traditional structure of dual mode bandpass filter is formed by a circular or rectangular resonator, which causes excessive circuit areas to be occupied. A 5.2 GHz bandpass filter, based on the triple-branch self-coupled ring resonator, is designed to achieve most essential features such as low insertion loss, compact size, and wide stopband. The coupling of each coupled branch introduces the even-and odd-mode perturbation, which produce sharp bandpass and wide stopband responses due to the
more &raquo; ... generation of band-edge transmission zeros. The simulation and measurement in this paper are respectively verified by using IE3D electromagnetic simulator and Agilent's HP8722C network analyzer. Experimental results show that the filter has 19.2% bandwidth centered at 5.2 GHz, 1.2 dB insertion loss, 25 dB stopband rejection from 6 to 9 GHz. Moreover, the circuit size in this paper has been down to 9×6 mm 2 and can be applied to communication of microwave applications.
