NF-κB Transcription Factor p50 Critically Regulates Tissue Factor in Deep Vein Thrombosis

NF-B transcription factors regulate the expression of tissue factor (TF), a principal initiator of the coagulation cascade. Dominant among them is the p50/p65 heterodimer. Here we report that Andrographolide (Andro; a p50 inhibitor) and genetic deletion of p50 attenuated TF activity in stimulated endothelial cells and monocytes/macrophages. Results of the electrophoretic mobility "supershift" assay and chromatin immunoprecipitation demonstrated the direct interaction of the p50/p65 heterodimer
more » ... ith the NF-B site of the human TF promoter. Andro-treated and p50 null mice both exhibited blunted TF expression and reduced venous thrombosis, which were recapitulated by an anti-murine TF antibody in vivo. Our findings thus indicate that regulation of TF by NF-B transcription factor p50 is essential for the pathogenesis of deep vein thrombosis and suggest that specific inhibitors of p50, such as Andro, may be therapeutically valuable for preventing and perhaps treating venous thrombosis.

Evaluating entropy rate of high-dimensional chaos and shot noise from analog raw signals remains elusive and important in information security. We experimentally present an accurate assessment of entropy rate for physical process randomness. The entropy generation of optical-feedback laser chaos and physical randomness limit from shot noise are quantified and unambiguously discriminated using the growth rate of average permutation entropy value in memory time. more » ... of filtered laser chaos with varying embedding delay time is investigated experimentally and theoretically. High resolution maps of the entropy difference is observed over the range of the injection-feedback parameter space. We also clarify an inverse relationship between the entropy rate and time delay signature of laser chaos over a wide range of parameters. Compared to the original chaos, the time delay signature is suppressed up to 95% with the minimum of 0.015 via frequency-band extractor, and the experiment agrees well with the theory. Our system provides a commendable entropy evaluation and source for physical random number generation.

Parton energy loss in cold nuclei [article]

Within the generalized high-twist factorization formalism, we express the contribution from multiple parton scattering and induced gluon radiation to the DY dilepton spectra in terms of nuclear modified effective beam quark distribution function. We show that beam quark energy loss is characterized by jet transport parameter q̂, which is related to the local gluon density of the medium. Using the value of q̂ determined from the deeply inelastic scattering (DIS) data, we evaluate the nuclear
more » ... fication factor in Drell-Yan process in p+A collisions. Effects of parton energy loss in the DY spectra are found negligible in the Fermilab experimental data at E_ lab=800 GeV relative to parton shadowing while the predicted suppression of DY spectra are significant at E_ lab=120 GeV.

We have calculated the directed flow v_1 and charge-odd directed flow Δ v_1 for pions and protons in Au+Au collisions at √(s_NN)=200GeV by solving the coupled Boltzmann-Maxwell equations self-consistently. Our numerical results show that v_1 for pions and protons are all negative and have similar behavior and magnitude. In contrast we find a quite different behavior in Δ v_1 for pions and protons. The difference lies in that Δ v_1 for protons mainly comes from pressure gradients of the medium,
more » ... hile the dominant contribution to Δ v_1 for pions is from electromagnetic fields. Our results indicate that the effect of the electric field will slightly exceed that of the magnetic and lead to a small negative slope of Δ v_1 for pions. Our numerical results are consistent with experimental data and solve the sign puzzle in Δ v_1 for pions and protons in previous studies.
