Over recent decades large amounts of data about our Planet have become available. If this information could be easily discoverable, accessible and properly exploited, preserved and shared, it would potentially represent a wealth of information for a whole spectrum of stakeholders: from scientists and researchers to the highest level of decision and policy makers. By creating a Virtual Research Environment (VRE) using a service oriented architecture (SOA) tailored to the needs of Earth Science
more » ... S) communities, the EVER-EST (http://ever-est.eu) project provides a range of both generic and domain specific data analysis and management services to support a dynamic approach to collaborative research. EVER-EST provides the means to overcome existing barriers to sharing of Earth Science data and information allowing research teams to discover, access, share and process heterogeneous data, algorithms, results and experiences within and across their communities, including those domains beyond Earth Science. The main objective of this paper is to present the EVER-EST platform in all its components describing the most relevant use cases implemented by the Virtual Research Communities (VRCs) involved in the project. Albani et al: EVER-EST Art. 21, page 2 of 16 integration of innovative ICT components and state of art services for research data management in Earth Sciences. The E-infrastructure enables scientists to manage the entire research life cycle of their scientific investigations, attribute and credit findings, validate claims, preserve and share research materials and results with the scientific community and the general public. Central to this work is the use of Research Objects (ROs) as semantically rich aggregations of data, methods and people in scientific investigations supporting the implementation of FAIR guiding principles and the systemic change of science practices to Open Science, particularly in e-Science. ROs allow encapsulating scientific knowledge and provide a mechanism for preserving, sharing and discovering assets of reusable and reproducible research.

BACKGROUND. Cardiovascular disease occurs at lower incidence in premenopausal females compared with age-matched males. This variation may be linked to sex differences in inflammation. We prospectively investigated whether inflammation and components of the inflammatory response are altered in females compared with males. METHODS. We performed 2 clinical studies in healthy volunteers. In 12 men and 12 women, we assessed systemic inflammatory markers and vascular function using brachial artery
more » ... w-mediated dilation (FMD). In a further 8 volunteers of each sex, we assessed FMD response to glyceryl trinitrate (GTN) at baseline and at 8 hours and 32 hours after typhoid vaccine. In a separate study in 16 men and 16 women, we measured inflammatory exudate mediators and cellular recruitment in cantharidin-induced skin blisters at 24 and 72 hours. RESULTS. Typhoid vaccine induced mild systemic inflammation at 8 hours, reflected by increased white cell count in both sexes. Although neutrophil numbers at baseline and 8 hours were greater in females, the neutrophils were less activated. Systemic inflammation caused a decrease in FMD in males, but an increase in females, at 8 hours. In contrast, GTN response was not altered in either sex after vaccine. At 24 hours, cantharidin formed blisters of similar volume in both sexes; however, at 72 hours, blisters had only resolved in females. Monocyte and leukocyte counts were reduced, and the activation state of all major leukocytes was lower, in blisters of females. This was associated with enhanced levels of the resolving lipids, particularly D-resolvin. CONCLUSIONS. Our findings suggest that female sex protects against systemic inflammation-induced endothelial dysfunction. This effect is likely due to accelerated resolution of inflammation compared with males, specifically via neutrophils, mediated by an elevation of the D-resolvin pathway. TRIAL REGISTRATION. Data-intensive science communities are progressively adopting FAIR practices that enhance the visibility of scientific breakthroughs and enable reuse. At the core of this movement, research objects contain and describe scientific information and resources in a way compliant with the FAIR principles and sustain the development of key infrastructure and tools. This paper provides an account of the challenges, experiences and solutions involved in the adoption of FAIR around research objects over
more » ... everal Earth Science disciplines. During this journey, our work has been comprehensive, with outcomes including: an extended research object model adapted to the needs of earth scientists; the provisioning of digital object identifiers (DOI) to enable persistent identification and to give due credit to authors; the generation of content-based, semantically rich, research object metadata through natural language processing, enhancing visibility and reuse through recommendation systems and third-party search engines; and various types of checklists that provide a compact representation of research object quality as a key enabler of scientific reuse. All these results have been integrated in ROHub, a platform that provides research object management functionality to a wealth of applications and interfaces across different scientific communities. To monitor and quantify the community uptake of research objects, we have defined indicators and obtained measures via ROHub that are also discussed herein.

The novel coronavirus disease (COVID-19) outbreak, caused by SARS-CoV-2, represents the greatest medical challenge in decades. We provide a comprehensive review of the clinical course of COVID-19, its comorbidities, and mechanistic considerations for future therapies. While COVID-19 primarily affects the lungs, causing interstitial pneumonitis and severe acute respiratory distress syndrome (ARDS), it also affects multiple organs, particularly the cardiovascular system. Risk of severe infection
more » ... nd mortality increase with advancing age and male sex. Mortality is increased by comorbidities: cardiovascular disease, hypertension, diabetes, chronic pulmonary disease, and cancer. The most common complications include arrhythmia (atrial fibrillation, ventricular tachyarrhythmia, and ventricular fibrillation), cardiac injury [elevated highly sensitive troponin I (hs-cTnI) and creatine kinase (CK) levels], fulminant myocarditis, heart failure, pulmonary embolism, and disseminated intravascular coagulation (DIC). Mechanistically, SARS-CoV-2, following proteolytic cleavage of its S protein by a serine protease, binds to the transmembrane angiotensin-converting enzyme 2 (ACE2) -a homologue of ACE-to enter type 2 pneumocytes, macrophages, perivascular pericytes, and cardiomyocytes. This may lead to myocardial dysfunction and damage, endothelial dysfunction, microvascular dysfunction, plaque instability, and myocardial infarction (MI). While ACE2 is essential for viral invasion, there is no evidence that ACE inhibitors or angiotensin receptor blockers (ARBs) worsen prognosis. Hence, patients should not discontinue their use. Moreover, renin-angiotensin-aldosterone system (RAAS) inhibitors might be beneficial in COVID-19. Initial immune and inflammatory responses induce a severe cytokine storm [interleukin (IL)-6, IL-7, IL-22, IL-17, etc.] during the rapid progression phase of COVID-19. Early evaluation and continued monitoring of cardiac damage (cTnI and NT-proBNP) and coagulation (D-dimer) after hospitalization may identify patients with cardiac injury and predict COVID-19 complications. Preventive measures (social distancing and social isolation) also increase cardiovascular risk. Cardiovascular considerations of therapies currently used, including remdesivir, chloroquine, hydroxychloroquine, tocilizumab, ribavirin, interferons, and lopinavir/ritonavir, as well as experimental therapies, such as human recombinant ACE2 (rhACE2), are discussed.
