MicroRNA 27b promotes cardiac fibrosis by targeting the FBW7/Snail pathway

Our study aspires to understand the impact of miR-27b on myocardial fibrosis as well as its functional mechanism. 12 days post the ligation of coronary artery in rats, the expression of miR-27b in the peri-infarction region was elevated. Treating cultivated rat neonatal cardiac fibroblasts (CFs) with angiotensin II (AngII) also enhanced the miR-27b expression. FBW7 was found to be the miR-27b's target since the overexpression of miR-27b reduced the transcriptional level of FBW7. The enhanced expression of FBW7 protein abrogated the effects of miR-27b in cultured CFs, while the siRNA silence of FBW7 promoted the pro-fibrosis activity of AngII. As to the mechanism, we found that the expression of FBW7 led to the degradation of Snail, which is an important regulator of cardiac epithelial-mesenchymal transitions. Importantly, inhibition of miR-27b abrogated the coronary artery ligation (CAL) induced cardiac fibrosis in vivo, suggesting that it might be a potential target for the treatment of fibrosis associated cardiac diseases.

As one of the most important types of (weaker) supervised information in machine learning and pattern recognition, pairwise constraint, which specifies whether a pair of data points occur together, has recently received significant attention, especially the problem of pairwise constraint propagation. There are two underlying motivations for us to write this survey paper: the first is to provide an up-to-date review of the existing literature, and the second is to offer some insights into the studies of pairwise constraint propagation. To provide a comprehensive survey, we not only categorize existing propagation techniques but also present detailed descriptions of representative methods within each category.

Scutellaria baicalensis Inhibits Coxsackievirus B3-Induced Myocarditis Via AKT and p38 Pathways

Scutellaria baicalensis Georgi has been widely used in China for treatment of various diseases. This study investigated the effect of Scutellaria baicalensis Georgi extracts (SBE) against Coxsackievirus B3 (CVB3)-induced myocarditis in vitro and in vivo. In vitro, Hela cells and primary myocardial cells were infected with CVB3 and treated with SBE (50-800 µg/ml) and ribavirin (200 µM) for 48 h and then determined by CCK8 assay. more » ... myocarditis model was induced in male BALB/c mice by injecting CVB3 suspension intraperitoneally for three times, followed by treatment with SBE (400 and 200 mg/kg) and ribavirin (100 mg/kg) for 28 days. SBE ameliorated the cytotoxicity of CVB3 in Hela cells, especially at 400 µg/ml (39.93% vs 65.67%, p < 0.05) without influencing cell growth and also significantly reduced CVB3 replication in primary myocardial cells. The levels of AKT, ERK, and p38 were increased after CVB3 infection. SBE could downregulate the expressions of AKT and p38. In vivo, the mortality rate from CVB3 reached to 66.67%, while 10.00% and 23.33% of this came after 400 and 200 mg/kg SBE treatment, respectively (p < 0.05). The CVB3 replication was obviously reduced after SBE administration from day 5. Similarly, the levels of AKT, ERK, and p38 mRNAs and proteins were increased, and SBE suppressed the expression of AKT and p38. Our study indicates that SBE is a promising potent antiviral agent against CVB3-induced myocarditis by inhibition of virus replication via depressing AKT and p38 expressions.

and purpose: Spinal cord injury (SCI) involves serious damage that can result in abnormal or absent motor and sensory functions and a disruption of autonomic function, and a series of pathological reactions occur after the injury. As a type of small non-coding RNA, microRNAs (miRNAs) have been verified to inhibit gene expression via post-transcriptional regulation. This review mainly focuses on recent advances regarding the roles of miRNAs following SCI. Methods: The Preferred Reporting Items
more » ... r Systematic Reviews and Meta-Analyses guidelines were adopted. The studies regarding the roles of miRNAs following SCI were identified through PubMed, Embase and Web of Science. We summarise the changes in expression levels of miRNAs and discuss the roles of miRNAs after SCI. Results: A total of 77 empirical studies meeting the inclusion criteria were identified. Existing studies showed that miRNAs were temporally altered and had effects on apoptosis, inflammation, angiogenesis, astrogliosis, oligodendrocyte development, axonal regeneration and remyelination after SCI. The alteration of miRNAs and the regulative action of pathological reactions can also provide opportunities for potential therapeutic interventions. "miRNA replacement therapy" aims to transfer miRNAs into diseased cells via delivery techniques and improve targeting effectiveness in cells, and this novel therapeutic tool provides a promising technique to promote the repair of SCI and reduces functional deficits. Conclusions: This review is helpful for understanding the underlying mechanisms of SCI and the potential clinical value of miRNAs. miRNAs have the potential to be attractive tools and targets for novel diagnostic and therapeutic approaches of SCI.

Application of a New Geophone and Geometry in Tunnel Seismic Detection

Seismic imaging is the most effective geophysical method and has been extensively implemented to detect potential geological hazards in tunnels during construction. The coupling of geophones and the design of geometry in tunnels are the two major challenges. more » ... ion occurs. This semi-automatic coupling design within the geophone not only guarantees good coupling, but reduces the time and costs usually required to install a traditional geophone. In the use of geophones for tunnel seismic detection, we propose two new two-dimensional (2D) seismic geometries based on the two commonly used geometries. A test to assess the effectiveness of the qualities of imaging from four geometries was completed by comparing the results of the forward modeling of sandwich models. The conclusion is that the larger the horizontal offset of the layout geometry, the higher the resolution of the imaging; the larger the vertical offset, the weaker the mirror image. The vertical offset is limited due to the narrow tunnel condition. Therefore, the mirror effect cannot be entirely eliminated; however, it can be further suppressed by constructing 2D geometry. The two newly proposed 2D geometries caused the imaging arc of the inter-layer, but suppressed the mirror image. The mirror image added a significant number of errors to the data, which could misguide tunnel construction; therefore the new 2D geometries are more reasonable than the two most commonly used. We applied one of the two new 2D geometries that was more practical to an actual project, the Chongqing Jinyunshan Tunnel in China, and acquired high-quality seismic data using two semi-automatic coupling geophones. The detection results were essentially consistent with the excavation conclusions.
