Urinary metabolomic investigations in vitiligo patients

Urinary metabolomics is a useful non-invasive tool for large-scale screening of disease-related metabolites. However, no comprehensive urinary metabolomic analysis of vitiligo is presently available. To investigate the urine metabolic pattern of vitiligo patients, we conducted a combined cross-sectional and prospective self-control cohort study and an untargeted urinary metabolomic analysis. more » ... and 71 differential metabolites between two groups were identified. Pathway enrichment analysis revealed that drug metabolism-cytochrome P450, biopterin metabolism, vitamin B9 (folate) metabolism, selenoamino acid metabolism, and methionine and cysteine metabolism showed significant enrichment in vitiligo patients compared with the status in healthy controls. In the self-control cohort, 46 active vitiligo patients were recruited to analyse the urinary metabolic signatures after treatment. All of these patients were asked to undertake follow-up visits every 2 months three times after first consulting and the disease stage was evaluated compared with that at the last visit. Folate metabolism, linoleate metabolism, leukotriene metabolism, alkaloid biosynthesis, and tyrosine metabolism were predicted to be involved in vitiligo activity. Our study is the first attempt to reveal urinary metabolic signatures of vitiligo patients and provides new insights into the metabolic mechanisms of vitiligo.

This paper introduces atomic force microscope (AFM) deposition method to fabricate nanostructures and nanodevices. Field emission theory is introduced in this paper, which provides theoretical explanation for AFM deposition. Dot matrixes are fabricated by AFM deposition on three different substrates, Si, Au and GaAs. Differences of deposition on the three substrates are discussed. Tegaserod Maleate Inhibits Esophageal Squamous Cell Carcinoma Proliferation by Suppressing the Peroxisome Pathway

Esophageal squamous cell carcinoma (ESCC) and esophageal adenocarcinoma (EAC) are the two major types of esophageal cancer (EC). ESCC accounts for 90% of EC. Recurrence after primary treatment is the main reason for poor survival. Therefore, recurrence prevention is a promising strategy for extending the 5-year survival rate. more » ... ignaling pathway, in which the key molecules peroxisome membrane protein 11B (PEX11B) and peroxisome membrane protein 13 (PEX13) were downregulated. The immunofluorescence, catalase activity assay, and reactive oxygen species (ROS) confirmed that downregulation of these proteins was related to impaired peroxisome function. Furthermore, we found that PEX11B and PEX13 were highly expressed in ESCC, and knockout of PEX11B and PEX13 further demonstrated the antitumor effect of tegaserod maleate. Importantly, tegaserod maleate repressed ESCC tumor growth in a patient-derived xenograft (PDX) model in vivo. Our findings conclusively demonstrated that tegaserod maleate inhibits the proliferation of ESCC by suppressing the peroxisome pathway.

Two novel 3-nor-methyl-chamigrane sesquiterpene peroxides, named steperoxide A (1) and B (2), have been isolated from basidiomycete Steccherinum ochraceum. This is the first report on the isolation of chamigrane sesquiterpene from higher fungi. The structures of 1 and 2 were established on the basis of spectroscopic data and single-crystal X-ray analysis. Mushrooms have proved to be a rich source of secondary metabolites with unusual structures as well as interesting biological activities.
more » ... te their potential for drug development, few bioactive metabolites have been reported from mushrooms as compared with higher plants and microbes. Over the last 10 years, our main research focus has been secondary metabolites from the untapped resources of higher fungi found in China. 1-5 As a continuation of our studies on structurally interesting and biologically active natural products from higher fungi, chemical investigation on fermentation broth of Steccherinum ochraceum has resulted in the isolation of two rare and novel chamigrane norsesquiterpene peroxides, named steperoxides A (1) and B (2). So far, almost all chamigrane sesquiterpenoids were isolated from marine creatures, and most of them contained at least one halogen. 6-12 This is the first report on the isolation of chamigrane sesquiterpene from higher fungi. We present herein the structural elucidation of these two compounds by spectroscopic data in conjunction with singlecrystal X-ray analysis. Steperoxide A (1) 13 was obtained as colorless crystals (in MeOH) with a molecular formula of C 14 H 22 O 4 as established by HREIMS at m/z 254.1511 [M] + (calcd for 254.1513), requiring 4 degrees of unsaturation. The IR absorptions revealed the presence of hydroxyl (3502 cm À1 ) and carbonyl (1707 cm À1 ) functionalities. The 1 H NMR spectrum (Table 1) showed the presence of three methyls (d H 1.86, 1.20, and 0.93) attached to quaternary carbons, and two oxymethine groups (d H 4.12 and 3.71). The 13 C NMR and HSQC spectra (Table 1) resolved 14 carbon resonances comprising one carbonyl group at d C 208.1, two oxygenated methine carbons at d C 80.2 and 70.1, one oxygenated quaternary carbon at d C 89.6, as well as three methyls (d C 27.2, 25.0, and 23.9), five methylenes (d C 36.6, 35.2, 32.6, 28.0, 25.9), and two quaternary carbons (d C 42.7 and 35.7). Accordingly, a three-ring structure was required for 1 to fulfill the unsaturation requirement.

Sparse R-CNN is a recent strong object detection baseline by set prediction on sparse, learnable proposal boxes and proposal features. In this work, we propose to improve Sparse R-CNN with two dynamic designs. First, Sparse R-CNN adopts a one-to-one label assignment scheme, where the Hungarian algorithm is applied to match only one positive sample for each ground truth. Such one-to-one assignment may not be optimal for the matching between the learned proposal boxes and ground truths. To
more » ... this problem, we propose dynamic label assignment (DLA) based on the optimal transport algorithm to assign increasing positive samples in the iterative training stages of Sparse R-CNN. We constrain the matching to be gradually looser in the sequential stages as the later stage produces the refined proposals with improved precision. Second, the learned proposal boxes and features remain fixed for different images in the inference process of Sparse R-CNN. Motivated by dynamic convolution, we propose dynamic proposal generation (DPG) to assemble multiple proposal experts dynamically for providing better initial proposal boxes and features for the consecutive training stages. DPG thereby can derive sample-dependent proposal boxes and features for inference. Experiments demonstrate that our method, named Dynamic Sparse R-CNN, can boost the strong Sparse R-CNN baseline with different backbones for object detection. Particularly, Dynamic Sparse R-CNN reaches the state-of-the-art 47.2% AP on the COCO 2017 validation set, surpassing Sparse R-CNN by 2.2% AP with the same ResNet-50 backbone.

Acid Modification of the Unsupported NiMo Catalysts by Y-Zeolite Nanoclusters

Unsupported NiMo catalyst has high hydrogenation activity due to its high active site distribution. However, low specific surface area and pore distribution greatly limit the efficient utilization of the active components. The Y-zeolite nanoclusters were hydrothermally synthesized and introduced into the unsupported NiMo catalysts from a layered nickel molybdate complex oxide. The XRD, N2 adsorption-desorption, FT-IR, Py-IR, SEM, NH3-TPD, and TEM were used to characterize all catalysts. The
more » ... nzothiophene (DBT) hydrodesulfurization (HDS) reaction was performed in a continuous high pressure microreactor. The results showed that the specific surface area, pore volume, and average pore size of the unsupported NiMo catalysts were greatly increased by the Y-zeolite nanoclusters, and a more dispersed structure was produced. Furthermore, the Lewis acid and total acid content of the unsupported NiMo catalysts were greatly improved by the Y-zeolite nanoclusters. The HDS results showed that the unsupported NiMo catalysts modified by the nanoclusters had the same high desulfurization efficiency as the unmodified catalyst, but had more proportion of direct desulfurization (DDS) products. The results offer an alternative to reducing hydrogen consumption and save cost in the production of ultra clean diesel.
