Correction of squareness measurements of Vickers indenters due to the tilt of the pyramid axis

In Vickers hardness measurements, ISO 6507-2 and 6507-3 Standards require to verify that the quadrilateral of the pyramid indenter base has angles of 90° ± 0.2°. Such measurement is usually performed through optical measuring systems, which, rotating the diamond indenter, allows to evaluate the angles between two consecutive faces with high accuracy. These angles correspond to the angles of the quadrilateral base when the axis of the pyramid is perfectly perpendicular to the seating surface.
more » ... ertheless, when the pyramid axis is tilted, the angles between two consecutive faces are different from the corresponding angles on the quadrilateral base, thus a correction is required. In this work, a method to correct squareness measurements, based on a geometrical model, is presented.

Indentation modulus at macro-scale level measured from Brinell and Vickers indenters by using the primary hardness standard machine at INRiM

In this paper, the experimental procedure and calculation model for the measurement of the indentation modulus by using the primary hardness standard machine at INRiM in the macro-scale range at room temperature is described. The indentation modulus is calculated based on the Doerner-Nix linear model and from accurate measurements of indentation load, displacement, contact stiffness, and hardness indentation imaging. more » ... enters (Brinell test). Test force is provided by a dead-weight machine, and the occurring displacement is measured by a laser-interferometric system. The geometrical dimensions of both the Vickers and Brinell indentations are measured by means of a micro-mechanical system and optical microscopy imaging techniques. Applied force and indentation depth are measured simultaneously, at a 16 Hz sampling rate, and the resultant loading-unloading indentation curve is obtained. Preliminary tests are performed on metal and alloy samples. Considerations and comments on the accuracy of the proposed method and analysis are discussed.

In this work, the experimental method and the calculation model for the determination of indentation moduli, indentation work, and indentation creep of metallic materials, by means of macroscale-level forces provided by a primary hardness standard machine at the National Institute of Metrological Research (INRIM) at the at room temperature were described. more » ... imaging and from the slope of the indentation unloading curve by applying the Doerner-Nix linear model; indentation work, representing the mechanical work spent during the force application of the indentation procedure, was determined by calculating the areas under the loading–unloading indentation curve, through fitting experimental data with a polynomial law. Measurements were performed with a pyramidal indenter (Vickers test). The applied force was provided by a deadweight machine, and the related displacement was measured by a laser interferometric system. Applied forces and the occurring indentation depths were simultaneously measured: the resulting loading–unloading indentation curve was achieved. Illustrative tests were performed on metals and alloy samples. Discussion and comments on the suitability of the proposed method and analysis were reported.

Neuromodulatory action of picomolar extracellular Aβ42 oligomers on pre- and postsynaptic mechanisms underlying synaptic function and memory

Failure of anti-amyloid-β peptide (Aβ) therapies against Alzheimer's disease (AD), a neurodegenerative disorder characterized by high amounts of the peptide in the brain, raised the question of the physiological role of Aβ released at low concentrations in the healthy brain. To address this question, we studied the presynaptic and postsynaptic mechanisms underlying the neuromodulatory action of picomolar amounts of oligomeric Aβ42 (oAβ42) on synaptic glutamatergic function in male and female

We found that 200 pm oAβ42 induces an increase of frequency of miniature EPSCs and a decrease of paired pulse facilitation, associated with an increase in docked vesicle number, indicating that it augments neurotransmitter release at presynaptic level. oAβ42 also produced postsynaptic changes as shown by an increased length of postsynaptic density, accompanied by an increased expression of plasticity-related proteins such as cAMP-responsive element binding protein phosphorylated at Ser133, calcium-calmodulin-dependent kinase II phosphorylated at Thr286, and brain-derived neurotrophic factor, suggesting a role for Aβ in synaptic tagging. These changes resulted in the conversion of early into late long-term potentiation through the nitric oxide/cGMP/protein kinase G intracellular cascade consistent with a cGMP-dependent switch from short- to long-term memory observed in vivo after intrahippocampal administration of picomolar amounts of oAβ42 These effects were present upon extracellular but not intracellular application of the peptide and involved α7 nicotinic acetylcholine receptors. These observations clarified the physiological role of oAβ42 in synaptic function and memory formation providing solid fundamentals for investigating the pathological effects of high Aβ levels in the AD brains.SIGNIFICANCE STATEMENT High levels of oligomeric amyloid-β42 (oAβ42) induce synaptic dysfunction leading to memory impairment in Alzheimer's disease (AD). However, at picomolar concentrations, the peptide is needed to ensure long-term potentiation (LTP) and memory. Here, we show that extracellular 200 pm oAβ42 concentrations increase neurotransmitter release, number of docked vesicles, postsynaptic density length, and expression of plasticity-related proteins leading to the conversion of early LTP into late LTP and of short-term memory into long-term memory. These effects require α7 nicotinic acetylcholine receptors and are mediated through the nitric oxide/cGMP/protein kinase G pathway. The knowledge of Aβ function in the healthy brain might be useful to understand the causes leading to its increase and detrimental effect in AD.
