Characterization of Adult Rat Astrocyte Cultures

Astrocytes, a major class of glial cells, regulate neurotransmitter systems, synaptic processing, ion homeostasis, antioxidant defenses and energy metabolism. Astrocyte cultures derived from rodent brains have been extensively used to characterize astrocytes' biochemical, pharmacological and morphological properties. The aims of this study were to develop a protocol for routine preparation and to characterize a primary astrocyte culture from the brains of adult (90 days old) Wistar rats. For
more » ... s we used enzymatic digestion (trypsin and papain) and mechanical dissociation. Medium exchange occurred from 24 h after obtaining a culture and after, twice a week up to reach the confluence (around the 4 th to 5 th week). Under basal conditions, adult astrocytes presented a polygonal to fusiform and flat morphology. Furthermore, approximately 95% the cells were positive for the main glial markers, including GFAP, glutamate transporters, glutamine synthetase and S100B. Moreover, the astrocytes were able to take up glucose and glutamate. Adult astrocytes were also able to respond to acute H 2 O 2 exposure, which led to an increase in reactive oxygen species (ROS) levels and a decrease in glutamate uptake. The antioxidant compound resveratrol was able to protect adult astrocytes from oxidative damage. A response of adult astrocytes to an inflammatory stimulus with LPS was also observed. Changes in the actin cytoskeleton were induced in stimulated astrocytes, most likely by a mechanism dependent on MAPK and Rho A signaling pathways. Taken together, these findings indicate that the culture model described in this study exhibits the biochemical and physiological properties of astrocytes and may be useful for elucidating the mechanisms related to the adult brain, exploring changes between neonatal and adult astrocytes, as well as investigating compounds involved in cytotoxicity and cytoprotection.

The effect of different physical therapy procedures in ultramarathons

Aims: To analyze the effects of physical therapy procedures in reducing acute pain in runners. A second aim was to investigate the impact of weather conditions on the number of physical therapy visits during the Ultramarathon Rio24 h. Methods: This retrospective study was performed at the Physical Education Center Admiral Adalberto Nunes (CEFAN). Medical records were analyzed. more » ... n Rio24 h. The variation in the Visual Analog Scale score (VAS) before and after each treatment were analyzed. After the evaluation procedure, another professional had decided the best procedure for the assessed athlete: massotherapy, cryotherapy by immersion, or stretching. An ANOVA was performed to assess which procedures produce pain reduction immediately after the procedure. Results: A total of 1,995 care services (228 in women) were performed in 602 athletes (84 women). Analyzing the treatments that used only one modality (n = 512 visits), the final pain decreased significantly over the initial pain for the three treatments. It was verified that massotherapy, cryotherapy, and stretching promote significant intragroup differences, with no intergroup differences. The environmental conditions were not associated with the number of visits. Conclusion: Massotherapy, cryotherapy, and stretching are effective treatments for acute pain reduction during ultramarathons. The weather conditions are not associated with the number of physical therapy visits.

Balanced Interval Coloring [article]

We consider the discrepancy problem of coloring n intervals with k colors such that at each point on the line, the maximal difference between the number of intervals of any two colors is minimal. Somewhat surprisingly, a coloring with maximal difference at most one always exists. Furthermore, we give an algorithm with running time O(n n + kn k) for its construction. This is in particular interesting because many known results for discrepancy problems are non-constructive. This problem naturally
more » ... models a load balancing scenario, where n tasks with given start- and endtimes have to be distributed among k servers. Our results imply that this can be done ideally balanced. When generalizing to d-dimensional boxes (instead of intervals), a solution with difference at most one is not always possible. We show that for any d > 2 and any k > 2 it is NP-complete to decide if such a solution exists, which implies also NP-hardness of the respective minimization problem. In an online scenario, where intervals arrive over time and the color has to be decided upon arrival, the maximal difference in the size of color classes can become arbitrarily high for any online algorithm.

The automorphism group of F_p induces an action of GL_2(p) on the three-dimensional subspaces of Z(F_p) ( Z_p)^4. The orbits of this action are in one-to-one correspondence with the isomorphism classes of loops from A_p. We describe the orbits, and hence we classify the loops of A_p up to isomorphism. It is known that every commutative automorphic p-loop is nilpotent when p is odd, and that there is a unique commutative automorphic loop of order 8 with trivial center. Knowing A_p up to isomorphism, we easily obtain a classification of commutative automorphic loops of order p^3. There are precisely 7 commutative automorphic loops of order p^3 for every prime p, including the 3 abelian groups of order p^3.

Persistence may be defined as high sustained yield over multi-harvest. Genetic insights about persistence are essential to ensure the success of breeding programs and any biomass-based project. This paper focuses on assessing the biomass yield persistence for bioenergy purpose of 100 elephantgrass clones measured in six growth seasons in Brazil. To assess the clones' persistence, an index based on random regression models and genotype-ideotype distance was proposed. Results suggested the
more » ... ce of wide genetic variability between elephantgrass clones, and that the yield trajectories along the harvests generate genetic insights into elephantgrass clones' persistence and G x E interaction. A gene pool that acts over the biomass yield (regardless of the harvest) was detected, as well as other gene pools, which show differences on genes expression (these genes are the major responsible for clones' persistence). The lower and higher clones' persistence was discussed based on genome dosage effect and natural biological nitrogen fixation ability applied to bioenergy industry. The huge potential of energy crops necessarily is associated with genetic insights into persistence, so just this way, breeding programs could breed a new cultivar that fulfills the bioenergy industries.

Abstract We investigate the teleportation of a qubit from a comoving sender Alice to an inertial receiver Bob in an asymptotically flat Robertson-Walker expanding spacetime. We use scalar or fermionic field modes inside Alice's and Bob's ideal cavities and show the degradation of the teleportation quality, as measured by the fidelity, through a mechanism governed by spacetime expansion. This reduction is demonstrated to increase with the rapidity of the expansion and to be highly sensitive to the coupling of the field to spacetime curvature, becoming considerably stronger as it reduces from conformal to minimal. We explore a perturbative approach in the cosmological parameters to compute the Bogoliubov coefficients in order to evaluate and compare the fidelity degradation of fermionic and scalar fields. *

A Constructive Proof of the Cycle Double Cover Conjecture [article]

The cycle double cover conjecture states that a graph is bridge-free if and only if there is a family of edge-simple cycles such that each edge is contained in exactly two of them. It was formulated independently by Szekeres (1973) and Seymour (1979). In this paper, we settle the conjecture in the affirmative. In particular, we give an algorithm, which inductively constructs a cycle double cover in polynomial time.
