Determination of voriconazole in human plasma by liquid chromatography-tandem mass spectrometry

Abstract
A rapid and sensitive method for the determination of voriconazole in human plasma was developed. Voriconazole and the internal standard were extracted from plasma samples by liquid-liquid extraction with 2 ml of diethyl ether: dichloromethane (60: 40, v/v). The chromatographic separation was accomplished isocratically on a 150×4.6 mm, 5 m Zorbax extend C18 column at a flow rate of 0.7 mL/min. Detection by electrospray positive ionization mass spectrometry in the multiple-reaction monitoring mode was completed within 3.2 min. Linearity was over the concentration range 20-2000 ng/ml with a limit of detection of 5 ng/ml. Intra-and inter-day precision measured as relative standard deviation was <4.40% and <5.44%, respectively. The method was applied in a bioequivalence study of two tablet formulations of voriconazole.
